TIME KILL ASSAY EKSTRAK ETANOL BUNGA KECOMBRANG (Etlingera elatior) TERHADAP BAKTERI Streptococcus sanguinis ATCC 10556 (Kajian in vitro)

Streptococcus sanguinis is a commensal bacteria that has the role as a pioneering colonizer in the formation of biofilms in oral cavity and can be pathogenic when entering the bloodstream. Torch ginger flower extract (Etlingera elatior) is known to have antibacterial potential containing compounds such as flavonoids, saponins, tannins, and alkaloids. This in vitro study aims to determine the effect of the exposure of torch ginger flower extract to S. sanguinis through time-kill assay method.
The subject used in this study was Streptococcus sanguinis ATCC 10556. The test group used in this study was phosphate buffer saline solution (negative control) and torch ginger flower extract with a final concentration of 1x MIC (6.25%) with time-points of 1 and 3 hours. The test continued with suspension inoculation on BHI-Agar media, incubation for 24 hours and the calculation of the number of growing colonies using standard plate count method.
The results of the one-way ANOVA test showed a significant mean difference between the extract group and the negative control group (p<0>Post-Hoc Games Howell test between the extract group and the control group at the exposure time of 1 and 3 hour. Based on the results of this study, it can be concluded that torch ginger flower extract with concentration of 6,25% has no effect on reducing the number of colonies of S. sanguinis ATCC 10556 within 3 hours of exposure.
