Studi Distribusi Residu Gula Alpha-D-Mannopyranosyl dan Alpha-D-glucopyranosyl pada Saluran Intestinal Sugar Glider (Petaurus breviceps)

Sugar glider (Petaurus breviceps) merupakan mamalia berkantong yang hidup di daerah pepohonan atau hutan-hutan yang berasal dari Australia, Papua Nugini, dan Indonesia. Sugar glider termasuk hewan omnivora yang biasanya memakan invertebrata, mamalia kecil, getah, nektar, dan bunga. Penelitian ini dilakukan untuk mengetahui residu gula glikokonjugat, khususnya alpha-D- mannopyranosyl sekaligus alpha-D-glucopyranosyl, pada saluran intestinal sugar glider. Sampel penelitian yang diambil adalah duodenum, jejunum, ileum, sekum, kolon, dan rektum dari dua ekor Petaurus breviceps jantan dewasa. Sampel difiksasi menggunakan Phosphate Buffer Saline formalin 10% dan jaringan diproses dengan metode parafin. Blok sampel dipotong setebal 8 μm. Pewarnaan yang digunakan adalah hematoksilin eosin (HE) dan lektin concanavalin A (ConA). Pengamatan dilakukan dengan mikroskop cahaya kemudian diambil gambarnya menggunakan OptiLab Viewer. Data yang diperoleh dianalisis secara deskriptif kualitatif berdasarkan intensitas warna cokelat akibat reaksi antara lektin yang digunakan dengan reseptor gula spesifik. Hasil menunjukkan sel Goblet di permukaan usus menunjukkan hasil negatif. Sel Goblet di kripte kolon menunjukkan hasil positif heterogen. Hasil positif heterogen didapat pada brush border jejunum dan rektum. Pola reaktivitas lektin ConA terhadap saluran intestinal Petaurus breviceps menunjukkan hasil beragam.

Sugar gliders (Petaurus breviceps) are marsupial mammal that live in areas of trees or forests originating from Australia, Papua New Guinea, and Indonesia. Sugar glider are omnivorous that usually eat invertebrates, small mammals, sap, ncktar, and flowers. This study was conducted to determine the residues glycoconjugate sugars, particularly alpha-D-mannopyranosyl as well as alpha-D-glucopyranosyl, in sintestinal tract of sugar glider. The samples taken were duodenum, jejunum, ileum, cecum, colon, and rectum of two adult males Petaurus breviceps. Samples were fixed using 10% formalin Phosphate Buffer Saline and tissues were processed using paraffin method. Sample blocks were cut to 8μm thickness. The stains used were hematoxylin eosin (HE) and lectin concanavalin A (ConA). Observations were made with a light microscope and images were taken using OptiLab Viewer. The data obtained were analyzed descriptively qualitatively based on the intensity of the brown color due to the reaction between the lectins used and specific sugar receptors. The results showed that Goblet cells on the surface of the intestines showed negative results. Goblet cells in colonic crypts showed heterogeneous positive results.
