Histomorfometri Uterus Tikus (Rattus norvegicus) Yang Diinjeksi Testosteron Dalam Usaha Membentuk Hewan Model Kista Ovari

Testosteron telah sering diteliti bahwa testosteron dapat menyebabkan perubahan metabolik dan dapat memengaruhi organ target jantan dan betina. Pada tubuh hewan betina, testosteron diubah menjadi estradiol oleh aksi aromatase. Kadar estradiol yang cukup tinggi dapat menyebabkan gangguan keseimbangan hormonal yang memicu terbentuknya kista ovarium. Hewan betina yang terkena kista ovarium menunjukkan gejala seperti perubahan ukuran dinding uterus. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian testosteron sebanyak 200 mg/kg BB pada ketebalan uterus tikus Rattus norvegicus. Penelitian dilakukan pada lima ekor tikus Rattus norvegicus galur Wistar, dua ekor tikus diinjeksikan testosteron sebanyak 200 mg/kg BB setiap hari selama 60 hari yang dibandingkan dengan tiga ekor tikus dengan injeksi NaCl fisiologis 0,1 ml sebagai kontrol. Analisis dilakukan dengan membandingkan rataan lebar uterus tikus kontrol dengan tikus injeksi testosteron. Pengolahan data secara statistik dilakukan dengan menggunakan SPSS Independent Sample T Test uji ANOVA. Hasil penelitian didapatkan nilai signifikansi 0,090 dengan P > 0,05 yang menunjukkan bahwa tidak ditemukannya perbedaan antara kelompok perlakuan yang diujikan. Ketebalan uterus tikus yang diinjeksikan testosteron sebanyak 200 mg/kg BB diperoleh hasil rataan ketebalan 746,48 ± 204,95 μm dan uterus tikus kontrol didapatkan rataan ketebalan 826,60 ± 444,38 μm. Kesimpulan penelitian ini menunjukkan bahwa pemberian testosteron 200 mg/kg berat badan selama 60 hari tidak memberikan pengaruh terhadap perubahan ketebalan uterus tikus Rattus norvegicus dalam usaha membentuk hewan model kista ovari.

Testosterone has been often studied that testosterone can cause metabolic changes and can affect male and female target organs. In female animal’s body, by aromatase actions, testosteron changed to be estradiol. Estradiol in high levels caused hormonal balance disorders that trigger the formation of ovary cyst. Female animals which affected by ovarian cysts show symptoms such as the changes of uterine wall size. This study aims to determine the effect of giving testosterone as much as 200 mg/kg weight on the uterine thickness of Rattus norvegicus rats. The study was conducted on five Rattus norvegicus Wistar strain. Two rats by injecting testosterone 200 mg/kg weight every day for 60 days compared with three rats with NaCl 0,1 ml injection as a control. Analysis was carried out by camparing the means of uterine width of control rats with testosterone injection rats. Statistical data was processed using SPSS Independent Sample T Test ANOVA. The results obtained a significance value of 0.090 with P > 0.05 which indicates that the two treatments has no differences found between the treatment groups tested. The thickness of the uterus of rats injected with testosterone as much as 200 mg/kg weight showed results with an average thickness of 746,48 ± 204,95 μm and uterine thickness of control rats showed results with an average thickness 826,60 ± 444,38 μm. The conclution of this study showed that the administration of testosterone 200 mg/kg weight in 60 days had no effect on uterine thickness of Rattus norvegicus rats in an attempt to form ovary cyst animal model.
