PENGARUH PENAMBAHAN EKSTRAK ETANOLIK KULIT MANGGIS TERHADAP DAYA ANTI JAMUR DAN PELEKATAN SOFT LINER PADA BASIS GIGI TIRUAN RESIN AKRILIK

2021 | Tesis-Spesialis | PROSTODONSIADenture stomatitis merupakan kondisi patologis yang sering terjadi pada mukosa yang tertutup gigi tiruan, utamanya disebabkan oleh infeksi Candida albicans. Soft liner adalah material pelapis gigi tiruan yang berfungsi sebagai bantalan lunak untuk mencegah kontak langsung antara basis gigi tiruan dengan jaringan mukosa yang terinflamasi. Penambahan agen anti jamur ke dalam material soft liner perlu dilakukan untuk mengetahui daya hambat pertumbuhan Candida albicans. Ekstrak kulit manggis (Garcinia mangostana Linn) diketahui memiliki daya antijamur. Tujuan penelitian ini adalah untuk mengetahui pengaruh penambahan ekstrak etanolik kulit manggis terhadap daya antijamur dan perlekatan soft liner pada basis gigi tiruan resin akrilik. Cara penelitian yaitu ekstrak etanolik kulit manggis dengan konsentrasi 5%, 10%, 20% dan 40% ditambahkan ke dalam soft liner, kemudian dievaluasi aktivitas antijamurnya terhadap Candida albicans dengan cara mengukur zona hambat pertumbuhan. Dua puluh lima pasang cakram akrilik dengan diameter 4,5 mm dan ketebalan 10 mm dipersiapkan, ekstrak etanolik kulit manggis dengan konsentrasi 5%, 10%, 20% dan 40% ditambahkan ke dalam soft liner dengan ketebalan 3 mm, lalu diletakkan diantara sepasang cakram akrilik. Uji kekuatan tarik menggunakan Universal Testing Machine dilakukan untuk mengevaluasi pelekatan soft liner dengan resin akrilik kuring panas. Data yang didapatkan dianalisis dengan uji Anava satu jalur. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan pada besarnya zona hambat pertumbuhan Candida albicans (p<0,05) antara kelompok kontrol dengan semua kelompok perlakuan. Terdapat perbedaan yang signifikan pada besarnya kekuatan tarik (p<0,05) antara kelompok kontrol dengan semua kelompok perlakuan. Kesimpulan penelitian ini adalah penambahan ekstrak etanolik kulit manggis ke dalam material soft liner berpengaruh meningkatkan daya antijamur soft liner terhadap Candida albicans namun menurunkan kekuatan tariknya terhadap basis gigi tiruan resin akrilik kuring panas walaupun masih memenuhi standar minimal penggunaan klinis.
Denture stomatitis is a common pathological condition affecting the mucosa under dentures, which is mainly caused by Candida albicans infection. Soft liner is a denture lining material that act as a soft cushion to prevent direct contact between denture base and inflamed mucosal tissue. The addition of various antimicrobial agents into soft liner material has been studied and obtained effective results inhibiting Candida albicans growth. Mangosteen (Garcinia mangostana Linn) pericarp extract is known to have antifungal activity and proven to inhibit Candida albicans growth. The aim of this study was to examine the antifungal activity and tensile strength between soft liner modified with mangosteen pericarp ethanolic extract and heat cured acrylic resin. The method of these study was the mangosteen pericarp ethanolic extract in various concentrations of 5%, 10%, 20% and 40% was added into soft liner, then evaluated its antifungal activity against Candida albicans by measuring the growth inhibition zone. Twenty fifth pairs of acrylic resin were prepared as experimental discs 10 mm in diameter and 4,5 mm in thickness. The mangosteen pericarp ethanolic extract in various concentrations of 5%, 10%, 20% and 40% was added into soft liner then added between acrylic resin disk with a thickness of 3 mm. Tensile strength test using a Universal Testing Machine is carried out to evaluate the bond strength between soft liner and heat cured acrylic resin. The data obtained were analyzed by one way Anava test. The results showed that there was a significant difference in the size of Candida albicans growth inhibition zones (p <0.05) between the control group and all treatment groups. There was statistically significant difference in tensile bond strength between control and case groups (p <0.05). The conclusion of this study is that the mangosteen pericarp ethanolic extract into soft liner material has an effect on increasing the soft liner antifungal properties of Candida albicans but decreasing its tensile strength toward heat cured acrylic resin denture base, although still qualify for minimal standard of clinical use
