HAEMATOLOGICAL FEATURER OF RAT AS ISCHEMIC STROKE ANIMAL MODEL

Penyumbatan mengurangi aliran darah dan oksigen ke otak, yang menyebabkan kerusakan atau kematian sel-sel otak. Salah satu model stroke yang paling relevan yang ditetapkan adalah tikus sebelum melanjutkan dengan penelitian pada manusia karena kesamaan patogenetik dari stroke pada tikus dan manusia. Tujuan dari penelitian ini adalah untuk mempelajari tentang gambar hematologis otak tikus betina yang diligasi di arteri karotid dan menyebabkan stroke pada tikus. Enam ekor tikus betina Wistar berusia tiga bulan digunakan dalam penelitian ini. Induksi stroke dilakukan dengan metode ligasi di arteri karotis komunis yang sinister. Tikus dipelihara selama satu minggu, setelah hari ketujuh tikus tersebut di-eutanasia dan kemudian organ otak diambil dan dicampurkan dengan larutan formalin persiapan darah hematologi. Parameter hematologis standar diukur atau dihitung dan data ditabulasi. Parameter hematologi dibandingkan dengan analisis varians (ANOVA). Hasil penelitian menunjukkan bahwa tikus yang diinduksi stroke ischemic mempunyai gambaran jumlah eritrosit, leukosut, limfosit serta trombosit yang lebih tinggi dibanding control tetapi jumlah neutrofil lebih rendah dibanding tikus kontrol.Berdasarkan hasil penelitian dapat disimpulkan bahwa induksi stroke ischemic dengan meligasi karotid komunis sinister selama 7 hari akan menyebabkan perubahan pada gambaran hematologis otak tikus betina dalam bentuk leukopenia, anemia, limfositopenia, trombositopenia dan neutrofilia.
Ischemic stroke is one of three types of stroke and occurs when an artery to the brain is blocked. The blockage reduces the blood flow and oxygen to the brain, leading to damage or death of brain cells. One of the most relevant stroke models that established are rodents before proceeding with the studies in human because of the pathogenetic similarities of strokes in rodents and humans. The aim of this research is to study about the brain haematological images of female rats that were ligated in their carotid arteries and caused stroke in rats. Six Wistar female rats of three months old were used in this research. Induction of stroke was done by ligation method in the carotid artery of communis sinister. The rats were kept for one week, after the seventh day the rats were euthanized and then the brain organ was removed and fixed with formalin solution for preparation of haematological blood. Standard haematological parameters were measured or calculated and data was tabulated. The hematological parameters were compared by the variance analysis (ANOVA). The results were the white blood cell, red blood cell, lymphocytes and mean platelet volume were higher in control rats but the neutrophil was lower in control rats. It can be concluded that the ligation of carotid of communis sinister for 7 days changes in the brain haematological images of female rats in the form of leukopenia, anemia, lymphocytopenia and also neutrophilia or neutrophilic leucocytosis.
