Pengaruh penambahan konsentrasi ekstrak jintan hitam (Nigella Sativa) pada soft liner terhadap daya anti jamur Candida albicans (Tinjauan penggunaan softliner pada obturator paska bedah kanker mulut)

Soft liner adalah material yang melapisi permukaan gigi tiruan yang berkontak dengan jaringan, berguna untuk perawatan jaringan dan menambah retensi pada obturator paska bedah kanker mulut. Kendala terbesar yang menyertai soft liner adalah pertumbuhan jamur seperti Candida albicans. Mikroba oral berperan penting dalam kanker sehingga perbaikan kebersihan mulut dapat membantu dalam membatasi perkembangan atau penyebaran kanker. Salah satu tanaman yang mempunyai daya anti jamur adalah Nigella sativa. Tujuan penelitian ini adalah untuk mengkaji pengaruh penambahan konsentrasi ekstrak Nigella sativa pada soft liner terhadap daya anti jamur Candida albicans. Sampel terdiri dari 27 disk soft liner dengan diameter 6 mm dan tebal 5 mm. Sampel dibagi menjadi 3 kelompok (n=9) yaitu kelompok I disk soft liner dengan ekstrak Nigella Sativa 5 %, kelompok II 10 % dan kelompok III 20 %. Daya anti jamur berupa zona hambat di sekitar disk soft liner diukur pada hari pertama dan hari kedua. Data yang diperoleh diuji dengan ANAVA dua jalur, dilanjutkan dengan uji post hoc LSD. Hasil penelitian menunjukkan rerata zona hambat tertinggi pada konsentrasi 20%, diikuti konsentrasi 10% dan terendah pada konsentrasi 5% baik untuk hari pertama maupun hari kedua. Rerata zona hambat hari kedua menurun dibandingkan hari pertama pada semua konsentrasi. Hasil ANAVA dua jalur dan uji post hoc LSD menunjukkan terdapat perbedaan bermakna antar kelompok konsentrasi (p<0,05) dan perbedaan tidak bermakna antar kelompok hari (p>0,05). Kesimpulan penelitian ini adalah terdapat pengaruh penambahan konsentrasi ekstrak jintan hitam (Nigella sativa) pada soft liner terhadap daya anti jamur Candida albicans, peningkatan konsentrasi menunjukkan peningkatan daya anti jamur dan terlihat aktivitas anti jamur bekerja pada hari pertama dan kedua.

Soft liners are materials that coat the tissue-bearing surface of a denture, useful for tissue care and improve retention of obturators after the surgery of oral cancer. The growth of fungi such as Candida albicans becomes the most serious shortcoming that accompanies the use of soft liners. Oral microbes contribute considerably to the growth of cancer, therefore improved oral hygiene will help inhibit the spread of cancer. One of the plants with antifungal activity is Nigella sativa. This study was aimed at investigating the effects of increased concentration of Nigella sativa extract on soft liners towards the antifungal activity against Candida albicans. The samples consist of 27 soft liner disks with a diameter of 6 mm and a thickness of 5 mm. Samples fall into 3 groups (n = 9), each of which comprises soft liner disk with Nigella Sativa extract by 5%, 10% and 20% for group I, II and III respectively. Antifungal activity in the form of inhibition zone around the soft liner disks was measured on the first day and the second day. The results were then analyzed with a two-way ANOVA, followed by a post-hoc LSD test. Results revealed the highest average inhibition zone at a concentration of 20%, followed by that of 10% and the lowest at 5% for both the first day and the second day. The inhibition zone of the second day decreased compared to that of the first day at all concentrations. The results of a two-way ANOVA and a post-hoc LSD test showed that there were significant differences between the concentration groups (p <0.05) and insignificant differences between groups of days (p> 0.05). The study concludes that there are effects of increased concentration of black cumin seeds (Nigella sativa) extract on soft liners towards the antifungal activity against Candida albicans, where the increased concentration shows higher antifungal activity and visible antifungal activities on the first and second day.
