Profil Hematologis Mencit (Mus Musculus Linnaeus, 1758) Galur Swiss dengan Induksi 7,12-Dimethylbenz[alpha]anthracene dan Paparan Medan Listrik Statis

2019 | Skripsi | S1 BIOLOGIMencit (Mus musculus) galur Swiss yang diinduksi menggunakan 7,12-Dimethylbenz[alpha]anthracene (DMBA) dapat digunakan sebagai model kanker payudara. Terapi kanker menggunakan medan listrik banyak dikembangkan karena bersifat noninvasif sehingga dapat meminimalisir efek samping. Electro-Capacitive Cancer Therapy (ECCT) adalah alat terapi kanker yang bekerja berdasarkan medan listrik statis untuk menghambat proliferasi sel kanker. Penggunaan ECCT perlu dievaluasi melalui pengujian profil hematologis karena dapat menggambarkan kondisi fisiologis individu. Penelitian ini bertujuan untuk mempelajari keamanan terapi medan listrik statis frekuensi menengah intensitas rendah terhadap profil hematologis mencit yang diinduksi DMBA. Hewan uji sebanyak 30 ekor dibagi menjadi 5 kelompok yaitu: tanpa perlakuan, noninduksi-nonterapi, noninduksi-terapi, induksi-nonterapi, dan induksi-terapi. Induksi kanker payudara dilakukan dengan cara pemberian DMBA per oral dosis 20 mg/kg bb sebanyak 10 kali selama 5 pekan. Terapi menggunakan kandang individu ECCT dimulai saat nodul tumor sudah dapat dipalpasi, berlangsung 10 jam per hari selama 21 hari. Sampel darah dikoleksi melalui intracardiac pada hari terakhir terapi, kemudian dilakukan pengukuran profil hematologis menggunakan hematology analyzer. Variabel yang dievaluasi meliputi: profil eritrosit (jumlah eritrosit, hemoglobin, hematokrit, MCV, MCH, MCHC), profil leukosit (jumlah leukosit total, persentase neutrofil dan limfosit, RNL) dan profil trombosit (jumlah trombosit, PCT, PDW, MPV). Data dianalisis secara statistik berdasarkan uji parametrik atau nonparametrik sesuai hasil uji normalitas. Hasil menunjukkan bahwa profil hematologis mencit yang diinduksi DMBA dan diterapi medan listrik statis berada dalam kisaran normal. Dapat disimpulkan bahwa terapi medan listrik statis pada mencit yang diinduksi DMBA relatif aman terhadap profil hematologis mencit.
Swiss mice (Mus musculus) induced by 7,12-Dimethylbenz[alpha]anthracene (DMBA) can be used as model for breast cancer. Cancer therapy with electric field has been developed as the method is noninvasive, thus minimizing side effects. Electro-Capacitive Cancer Therapy (ECCT), a device that works based on static electric field, is designed to inhibit cancer cell proliferation. The use of ECCT needs to be evaluated, one of which is through hematology test because it can reflects physiological conditions in individual. This research was conducted to study the safety of medium frequency and low intensity static electric field on hematological profile of DMBA-induced mice. Thirty animals were divided into 5 groups as untreated, noninduction-nontherapy, noninduction-therapy, induction-nontherapy, and induction-therapy. Breast cancer was induced by oral administration of DMBA dose 20 mg/kg bw for 10 times in 5 weeks. Therapy was started after nodules were palpable, lasting for total 10 hours/day in 21 consecutive days. Blood samples were collected via intracardiac on the last day of therapy, then hematological profile were assessed using hematology analyzer. Variables evaluated including: erythrocyte profile (total count, hemoglobin, hematocrit, MCH, MCV, MCHC), leukocyte profile (total count, percentage of neutrophil, percentage of lymphocyte, N/L), and platelet profile (total count, PCT, PDW, MPV). Data were statistically analyzed using parametric or nonparametric test based on normality test. Results demonstrated that hematological profile of mice after induced by DMBA and exposed to static electric field were within the normal range. In conclusion, therapy using static electric field in DMBA-induced mice is relatively safe.
