Pengaruh Lama Kontak Ekstrak Buah Belimbing Manis (Averrhoa carambola L.) Sebagai Larutan Kumur terhadap Pertumbuhan Streptococcus mutans

Plak gigi merupakan penyebab utama terjadinya karies gigi. Streptococcus mutans adalah bakteri penyebab terbentuknya plak gigi, yang dapat dihambat dengan senyawa antibakteri. Buah belimbing manis (Averrhoa carambola L.) memiliki senyawa antibakteri diantaranya flavonoid, tanin, dan saponin. Efek antibakteri dapat diperoleh dengan meningkatkan konsentrasi bahan antibakteri atau memperpanjang lama kontak bahan antibakteri. Penelitian ini bertujuan untuk mengetahui pengaruh lama kontak ekstrak buah belimbing manis (Averrhoa carambola L.) sebagai larutan kumur terhadap pertumbuhan Streptococcus mutans. Buah belimbing manis diperoleh dari perkebunan buah di Bojonegoro, Jawa Timur. Buah belimbing manis diekstraksi dengan metode maserasi menggunakan etanol 70%. Larutan kumur dibuat dengan komposisi sorbitol 0,01 mL, minyak permen 0,015 mL, ekstrak buah belimbing manis 0,25 g, dan 0,725 mL akuades. Streptococcus mutans dengan standar kekeruhan 0,5 McFarland dimasukkan 10 µL ke dalam conical tube, ditambahkan larutan kumur hingga bervolume 1 mL. Lama kontak yang digunakan yaitu 30 detik, 45 detik, dan 60 detik. Pertumbuhan bakteri dilakukan dengan menanamkan 10 µL suspensi bakteri yang telah dikontakkan dengan larutan kumur ke Plat Agar Darah dengan metode spread plate. Media tersebut diinkubasi dalam inkubator selama 24 jam pada suhu 37°C. Jumlah bakteri yang tumbuh dihitung menggunakan colony counter dengan satuan CFU/mL. Hasil penelitian diperoleh rerata jumlah koloni pada perlakuan lama kontak 30 detik, 45 detik, dan 60 detik secara berturut-turut adalah 14,75 x 10^2 CFU/mL, 7,25 x 10^2 CFU/mL, dan 2 x 10^2 CFU/mL. Hasil uji ANAVA satu jalur menunjukkan bahwa lama kontak ekstrak buah belimbing manis sebagai larutan kumur memiliki pengaruh yang bermakna terhadap pertumbuhan S. mutans (p<0,05). Hasil uji LSD0,05 menunjukkan bahwa terdapat perbedaan yang bermakna antar rerata jumlah koloni S. mutans. Kesimpulan penelitian ini, lama kontak ekstrak buah belimbing manis (A. carambola L.) sebagai larutan kumur berpengaruh terhadap pertumbuhan S. mutans.

Dental plaque is the main cause of dental caries. Streptococcus mutans is known as the cause of dental plaque formation, which can be inhibited by antibacterial compounds. Star fruit (Averrhoa carambola L.) has antibacterial compounds including flavonoid, tannin, and saponin. Antibacterial effects can be obtained by increasing the concentration of antibacterial ingredients or prolonging the exposure time of antibacterial ingredients. The aim of this research was to study the effect of exposure time of star fruit extract as a mouthwash on S. mutans growth. Star fruit was obtained from fruit plantations in Bojonegoro, East Java. It was extracted by maceration method with 70% ethanol. The composition of mouthwash was 0.01 mL sorbitol, 0.015 mL peppermint oil, 0.25 g star fruit extract, and 0.725 mL distilled water. Streptococcus mutans with 0.5 McFarland turbidity standard at amount of 10 μL was put into conical tube, a the mouthwash was added up to 1 mL. The exposure time used in this research was 30 seconds, 45 seconds, and 60 seconds. Bacterial growth was tested by inoculating 10 µL of bacterial suspension that had been exposed with mouthwash onto Blood Agar Plate by spread plate method. The media was incubated using incubator for 24 hours at 37°C. The number of bacteria was calculated using a colony counter in CFU/mL. The result showed the mean number of colonies after exposure time namely 30 seconds, 45 seconds and 60 seconds were 14.75 x 10^2 CFU/mL, 7.25 x 10^2 CFU/mL and 2 x 10^2 CFU/mL, respectively. The results of one-way ANOVA showed that among all three groups were significantly different (p<0.05). The LSD0,05 test results showed that there were significant differences within the groups of treatment. The conclusion was the exposure time of the star fruit extract as mouthwash influenced the growth of S. mutans.
