UJI AKTIVITAS ANTIALERGI FORMULA HERBAL IFP PADA TIKUS JANTAN WISTAR DENGAN MODEL HISTAMINE-INDUCED HIND PAW EDEMA

Penelitian ini merupakan salah satu bagian dari penelitian terintegrasi tentang uji praklinik Formula Herbal Imboost Flu-Pilek (IFP) yang bertujuan melihat aktivitas sinergis dari komponen kombinasi dalam menghambat reaksi alergi. Formula Herbal IFP merupakan kandidat fitofarmaka berbasis kombinasi ekstrak temulawak (Curcuma xanthorrhiza) 250 mg, ekstrak legundi (Vitex trifolia) 250 mg, ekstrak jahe (Zingiber officinale) 100 mg, ekstrak buah jeruk (Citrus sp.) 100 mg, dan ekstrak Echinacea purpurea 150mg. Penelitian ini mengacu pada metode Histamin-Induced Hind Paw Edema Test menggunakan tikus jantan galur Wistar sebagai hewan coba sebanyak 25 ekor yang dibagi kedalam 5 kelompok acak meliputi, 1 kelompok kontrol positif cetirizin (0,9 mg/kgBB), 1 kelompok kontrol negatif (pelarut CMC Na 0,5% 10 ml/kgBB), dan 3 kelompok variasi dosis Formula Herbal IFP (167,5 mg/kgBB; 335 mg/kgBB; dan 670 mg/kgBB). Parameter yang diamati adalah volume edema kaki tikus yang diukur dengan pletismometer setiap 30 menit selama 3 jam setelah injeksi histamin. Histamin 1% b/v diinjeksikan sebanyak 100 mikroL secara sub plantar pada kaki belakang kanan tikus sebagai induktor alergi. Data kuantitiatif yang diperoleh berupa nilai AUC (Area Under the Curve) dari kurva volume edema rata-rata terhadap waktu dan nilai persentase Daya Antialergi (%DAA). Dilakukan analisis terhadap data yang diperoleh menggunakan metode One Way ANOVA dengan taraf kepercayaan 95% dan dilanjutkan dengan uji LSD. Hasil penelitian ini menunjukkan bahwa peningkatan dosis Formula Herbal IFP berbanding lurus dengan potensinya sebagai antialergi. Dengan melakukan perbandingan terhadap kontrol positif, diperoleh persentase DAA dosis I, dosis II, dan dosis III berturut-turut senilai 0,46%; 0,60%; dan 0,86%.

This study was conducted as part of an integrated research regarding preclinical study of Herbal Formula Imboost Flu-Pilek (IFP), which aims to evaluate the synergistic activity of the combination components in inhibiting allergic reactions. Herbal Formula IFP refers to a fitofarmaka candidate that contains the combination of temulawak extract (Curcuma xanthorrhiza) 250 mg, legundi extract (Vitex trifolia) 250 mg, jahe extract (Zingiber officinale) 100 mg, orange extract (Citrus sp.) 100 mg, and Echinacea purpurea extract 150 mg. This study's method refers to Histamine-Induced Hind Paw Edema Test using 25 Wistar-strain male rats divided into 5 groups. The groups consist of: 1 group as positive control using cetirizine (0,9 mg/kgBB), 1 group as negative control (CMC-Na 0,5% 10 ml/kgBB), and 3 groups of Herbal Formula IFP with a varying doses (167,5 mg/kgBB; 335 mg/kgBB; and 670 mg/kgBB). The observed parameter is hind paw edema volume after histamine injection in every 30 minutes for 3 hours. One hundred microliter of histamine 1% w/v was injected via sub plantar route to ignite the allergic reaction. The quantitative data is available in a form of Area Under the Curve (AUC) from the plot of edema volume versus time and anti-allergic inhibition percentage (%DAA). Data collected in this study are analyzed using One Way ANOVA with 95% confidence rate and followed by the LSD test. The results of this study indicate a positive correlation between the dose of Herbal Formula IFP and its anti-allergic potency. With a comparison to positive control, the DAA persentage of dose I, dose II, and dose III consecutively are 0,46%; 0,60%; and 0,86%.
