EFEKTIVITAS OXYTETRACYLINE: TERHADAP Vibrio parahaemolyticus PADA KERAPU MACAN (Epinephelus juscogutattus)

Penelitian ini bertujuan untuk mengetabui efektivitas oxytetracycline terbadap Vibrio parahaen. Jlyticus pada ikan kerapu macan (Epinephe/us fuscoguttatus), yang dilakukan di Laboratorium Patologi Fakultas Kedokteran Hewan Universitas Gad jab Mada, Y ogyakarta. Penelitian dilakukan 2 tabap yaitu Pertama, uji pendahuluan meliputi pemumian isolat Vibrio parahaemolyticus, uji sensitivitas Vibrio parahaemolyticus terbadap antibiotik streptomycine, amoxycilline, enrojloxacine, oxytetracycline dan tetracycline dengan menggunakan metode Kirby-Bauer, penentuan nilai Minimum Inhibitory Concentration (MIC) antibiotik terpilib(oxytetracyc/ine) dengan Tube dilution test. Kedua, uji utama digunakan 70 ekor ikan kerapu macan dibagi 7 kelompok yang masing-masing kelompok berisi 10 ekor ikan. Satu kelompok tanpa infeksi Vibrio parahaemolyticus sebagai kontrol, sedan~kan 6 kelompok yang lain diinfeksi Vibrio parahaemolyticus konsentrasi 10 sel/ml secara rendaman sampai timbul gejala klinis, kemudian diberi antibiotik terpilib (oxytetracycline) pada 5 kelompok dengan dosis 1 x MIC, 2 x MIC, 4 x MIC, 6 x MIC dan 8 x MIC, sedangkan 1 kelompok tanpa pengobatan (kontrol +). Pengobatan secara rendaman 5 jam per bari berturut-turut selama 6 bari Vibrio parahaemolyticus sensitif terhadap oxytetracycline, tetracycline dan enrojloxacine. Oxytetracycline menunjukkan basil yang paling sensitif ( diameter zona terang 32 mm) dengan nilai MIC adalab 4 Jlg/ml. Daya kelangsungan bidup ikan yang diinfeksi Vibrio parahaemolyticus dan diobati dengan oxytetracycline secara rendaman selama 6 bari pada konsentrasi 24Jlg/ml (6 x MIC) sebesar 100% dengan gambaran perubaban bistopatologik yang lebib baik. Dari basil yang diperoleb maka dapat disimpulkan bahwa pemberian antibiotik oxytetracycline dengan konsentrasi 24Jlg/ml (6 x MIC) efektif dalam pengobatan ikan kerapu macan yang terinfeksi Vibrio parahaemolyticus secara rendaman selama 5 jam per bari selama 6 hari berturut-turut, Kata kunci : Kerapu macan (Epinephelus fuscoguttatus), V. parahaemo{vticus, oxytetracycline, efektivitas.

This research aimed to study effectiveness of oxytetracycline against Vibrio parahaemolyticus on tiger grouper (Epinephelus fuscoguttatus), done in Pathology. Laboratory of Veterinary Faculty, Gadjah Mada University in Yogyakarta. The research was carried out in two phased. First, introductory test included purification of isolate Vibrio parahaemolyticus, sensitivity test of Vibrio parahaemolyticus to antibiotics streptomycin, amoxicillin, enrojloxacine, oxytetracycline and tetracycline using Kirby Bauer method, and determination of minimum inhibitory Concentration (MIC) of selected antibiotics (oxytetracycline) using tube dilution test. Second, main test used 70 tiger grouper, divided into 7 groups with I 0 fishes each. One group was without infection Vibrio parahaemolyticus as control, while other six groups were infected with Vibrio parahaemolyticus, with concentration of I 06 celVml using submersion method until clinical symptom appeared. Then, five groups were providPd with selected antibiotic (oxytetracycline) with dosages of I x MIC, 2 x MIC, 4 x MIC, 6 x MIC, and 8 x MIC, respectively, while one other group was without medication (control +). Medication with submersion method was carried for 5 hours per day successively for 6 days. Vibrio parahaemolyticus was sensitive to oxytetracycline, tetracycline and enrojloxacine. Oxytetracycline indicated the most sensitive result (32 mm light zone diameter) with MIC score of 4J.Lg/ml. Survivability of fish infected with Vibrio parahaemolyticus and medicated with oxytetracycline with submersion method for six days in concentration of24 J.Lg/ml (6 x MIC) was IOO% with better histopathology change feature. The conclusion was that administration of 24 J.Lgiml (6 x MIC) oxytetracycline was effective in medication of tiger grouper infected with Vibrio parahaemolyticus in submersion for 5 hour per day for 6 days successively.
