Prosedur Pembuatan Preparat Histologi di National Laboratory Animal Center (NLAC) Thailand

Pembuatan preparat histologi yang baik dan tepat dapat digunakan untuk menunjang diagnosa penyakit (infeksi, peradangan, kanker) dan mengidentifikasi perubahan fungsi fisiologis tanpa tanda-tanda klinis. Teknik pembuatan preparat histologi yang disebut histoteknik dimulai dengan koleksi jaringan yang kemudian diproses sehingga menghasilkan preparat yang siap untuk diidentifikasi. Penulisan Tugas Akhir ini bertujuan untuk mengetahui langkah pembuatan preparat histologi menggunakan pewarnaan hematoxilyn-eosin dan mengevaluasi hasil. Pembuatan preparat dilakukan di National Laboratory Animal Center (NLAC), Thailand. Sampel yang digunakan berupa organ jantung, ginjal, hati dan pankreas. Proses pembuatan preparat menggunakan alat otomatis, sehingga hasil lebih cepat dan menghemat tenaga. Hasil dari pengamatan preparat histologi terdapat jaringan yang melipat, potongan tidak merata, pewarnaan tidak merata, dan adanya kotoran pada preparat histologi. Penyebab dari kerusakan preparat karena pisau kurang tajam, larutan pewarnaan yang sudah lama, air waterbath yang kotor. Kesimpulan dari pembuatan preparat histologi di NLAC selama periode Praktek Kerja Lapangan sudah cukup baik, namun perlu ada beberapa evaluasi dalam pemrosesan, misalnya penggunaan pisau mikrotom yang lebih tajam, menghomogenkan larutan pewarna sebelum proses staining, dan cara membersihkan air di dalam waterbath.

Manufacture a good slide histology can used to support the diagnosis of medical science (infection, inflammation and cancer) and in some circumstances these changes can be identified in the absence of obvious clinical sign and symptoms of diseade. Histoteknik is the technique of making histology preparation, begins with collect the sample which then processed until the preparat histology ready for identificatoin. The purpose of the study is to know procedure manufacture slide histology with the hematoxylin-eosin and can evaluate slide histology. Manufacture histology slide was made in National Laboratory Animal Center (NLAC). Heart, kidney, liver, and pancreas are use for sample. The procces manufacture of slide histology using automatic machine will be efficient. The result of observation slide histology there are folding, knife marks, stain precipitate, and dirty slide. Damage slide histology because the blade is not sharp, an old dye solution (hematoxylin-eosin) and dirty water in waterbath. Based on the making of histology preparations at NLAC during the field work practice period are good enough, but there needs to be some evaluation in processing, for example the use of sharper microtome blade, homogenizing the dye solution before the staining process and how to clean water in the waterbath.
