EVALUASI PELAKSANAAN STANDARD OPERATING PROCEDURE (SOP) ROSE BENGAL TEST (RBT) DI BALAI BESAR VETERINER WATES YOGYAKARTA

Rose Bengal Test (RBT) adalah uji aglutinasi sederhana dengan menggunakan antigen yang diwarnai dengan zat warna Rose Bengal. Pengujian ini dapat mendeteksi antibodi dari sampel susu, serum, cairan vagina dan semen. Tujuan penulisan Tugas Akhir ini adalah untuk membandingkan antara pelaksanaan pengujian dengan SOP RBT di BBVet Wates serta dengan standar OIE Terrestrial Manual 2016, EU Reference Laboratory dan Cromatest Linear Chemicals. SOP diperlukan sebagai pedoman dalam melaksanakan pengujian dan mengurangi tingkat kesalahan dan kelalaian yang mungkin dilakukan. Metode RBT di BBVet Wates Yogyakarta dilakukan dengan cara sampel (contoh : serum), kontrol positif dan negatif diambil masing-masing sebanyak 25 mikroliter diteteskan pada plat WHO dan dicampur dengan 25 mikroliter antigen Brucella, plat WHO digoyangkan selama 4 menit. Hasil positif diertimbangkan jika terlihat aglutinasi pada plat. Hasil negatif bila tidak terlihat aglutinasi. Hasil harus dibandingkan dengan kontrol. Prosedur RBT yang dilaksanakan di BBVet Wates telah sesuai dengan referensi.

The Rose Bengal Test (RBT) is a simple agglutination test for antibody detection against Brucella sp. Principally, in this test, the antibody would bind to Brucella antigen that coloured with Rose Bengal dye and forms agglutinate. The test could detect antibody from milk, serum, vaginal fluid and cement samples. The aim of this work is to compare the daily test application in BBVet Wates Yogyakarta with reference manual for RBT from the standard OIE Terrestrial Manual 2016, the EU Reference Laboratory and Cromatest Linear Chemicals RBT Manual. SOP is essentially required as a guideline to carry out testing and reduce the level of error and omission that might be done. The application of RBT method in BBVet Wates is described as follows, sample (e.g. serum), positive control dan negative control are taken as much as 25 microliter each and then dispense on the WHO plate and mix with 25 microliter RBT antigen, shake the WHO plate for 4 minutes. The result is considered positive when the agglutination appeared on the plate and for negative results, vice versa. The validity of the test could be seen from the performance of the controls and the comparison of the results and the controls. According to the observation result that has been done in this work, the application procedure of RBT conducted in BBVet Wates is consistent with the reference manuals.
