PENGARUH NANOFIBER EKSTRAK DAUN KELOR (Moringa oleifera Lam.) 0,1% TERHADAP PROLIFERASI OSTEOBLAS PADA PROSES OSTEOGENESIS (Kajian pada Osteoblast Cell Line MG63)

2018 | Tesis | MAGISTER ILMU KEDOKTERAN GIGI KLINISLatar Belakang: Nanofiber adalah sistem intra pocket drug delivery yang bersifat biokompatibel dan dapat dibuat menggunakan polimer kimia maupun herbal, seperti ekstrak tanaman kelor (Moringa oleifera Lam.). Kandungan β-carotene, protein, kalsium, kalium, dan kandungan zat antioksidan mempunyai efek osteoprotektif dan berpengaruh pada osteogenesis. Proses proliferasi osteoblas dalam osteogenesis merupakan salah satu proses penting. Sel osteoblas MG-63 merupakan sel line dengan sifat prototype dari sel osteoblas manusia dan telah banyak digunakan dalam pengujian biomaterial. Tujuan penelitian ini adalah untuk mengetahui pengaruh nanofiber dengan ekstrak daun kelor (Moringa oleifera Lam.) 0,1% terhadap proliferasi Osteoblast Cell Line MG63. Metode: setelah dibuat Nanofiber PVA-Kitosan-MOL, Nanofiber PVA-Kitosan dengan metode elektrospinning, dan ekstrak MOL 0,1% maka dilakukan penelitian. Kelompok yang diteliti adalah MOL 0,1%, Nanofiber PVA-Kitosan, dan Nanofiber MOL 0,1%. Pengamatan dilakukan pada inkubasi 24, 48, dan 72 jam. Pegamatan masing-masing 9 well per kelompok. Medium dibuang dan ditambahkan reagen MTT assay 100μL, setelah 4-6 jam diberikan stop solution sebanyak 100μL. Sel diinkubasi overnight kemudian dibaca menggunakan ELISA plate reader. Hasil dikonversi menjadi persentase proliferasi. Hasil: Proliferasi sel osteoblas MG63 meningkat pada inkubasi 24jam, 48jam, dan 72jam pada kelompok Nanofiber PVA-Kitosan-MOL 0,1%. Sedangkan nanofiber PVA-Kitosan dan ekstrak MOL 0,1% peningkatan hanya sampai 48 jam. Kesimpulan: Nanofiber ekstrak daun kelor (Moringa oleifera Lam.) 0,1 % meningkatkan proliferasi osteoblast sel line MG63.
Background: Intra pocket drug delivery system using nanofiber technologies is biocompatible materials which could be made by chemical or herbal polymers, such as Moringa oleifera Lam (MOL). Beta-carotene, protein, calcium, potassium, and antioxidant substances have osteoprotective effects which are beneficial for osteogenesis. The proliferation of osteoblasts in osteogenesis is an important process. MG-63 osteoblasts are cell lines with prototype properties of human osteoblast cells and have been widely used in biomaterials testing. The aim of this study was to determine the effect of nanofiber with 0.1% MOL extract on osteoblast proliferation in osteogenesis process (Study on Osteoblast Cell Line MG63). Methods: Nanofiber PVA-Kitosan-MOL and nanofiber PVA-Kitosan was made using electrospinning method. The subjects studied were MOL 0.1%, Nanofiber PVA-Chitosan, and Nanofiber MOL 0.1%. Observations were made 24, 48, and 72 hours incubation, seeded into 9 well for each groups. Medium were removed then MTT procedure were performed. The cells were incubated overnight, then read using an ELISA plate reader. Result: Proliferation of MG63 osteoblast cell line increased from incubation 24 hours, 48 hours, and 72 hours in nanofiber with 0.1% Moringa oleifera Lam extract. The highest proliferation of MG63 osteoblast cell line were incubated at 72 hours. The result were converted by proliferation percentage. Conclusion: Nanofiber with 0.1% Moringa leaf extract increased the proliferation of osteoblasts cell line MG63.
