TRANSESTERIFIKASI MINYAK JELANTAH DENGAN KATALIS LIPASE TERIMOBILISASI PADA KITOSAN BEAD; TRANSESTERIFICATION OF WASTE FRYING OIL USING LIPASE IMMOBILIZED ON CHITOSAN BEAD AS CATALYST

The study of activities of immobilized lipase on chitosan bead as catalyst for waste frying oil transesterification has been done. This study aimed to determine efficiency of lipase immobilization on chitosan bead and optimum mole oil:methanol ratio of the reaction, to know the activities from re-use lipase and to determine the value of Vmax and KM in the enzymatic reaction. Lipase immobilized was prepared by crosslinking method used glutaraldehyde with chitosan bead as supporting matrix. FAME concentration was determined by means of gas chromatography with internal standard. Efficiency of lipase immobilization on chitosan bead using cross-linking method has 56.44%. The highest conversion is obtained at condition of ratio mole oil:methanol 1:6 used lipase imobilized with 2.21% of FAME conversion. Repeated use of the free lipase at 2 cycles has decreased the FAME conversion by 42.97%, whereas the repeated use of lipase immobilized FAME conversion decreased 36.88% in the second cycle and 16.37% in the third cycle. The affinity of lipase immobilized was better than free lipase, seems to value of KM on immobilized lipase was 1.01 mg mL-1, smaller than free lipase, it was 1.07 mg mL-1.
