2015 | Skripsi | FMIPAIdentification and antibacterial activity test of ethyl acetate fractions of sponge Axinella sp. collected from Menjangan Island Sea, Bali Barat has been carried out. The isolation technique used in this study was meceration method, partition, and preparative thin layer chromatography (Preparative TLC). Maceration was performed by using dichloromethane:methanol (1:1) (v/v). The dichloromethane fraction obtained was then partitioned into ethyl acetate:water (1:1) (v/v). Ethyl acetate fraction was then concentrated to obtain the crude extract. The crude extract was separated by preparative thin layer chromatography by using mix eluents that was n-hexane:chloroform:ethyl acetate (4.0:7.5:7.5) (v/v/v). The separated fractions were then tested for antibacterial activity against E.coli. The fractions containing active compounds were analyzed by using LCMS (Liquid Chromatography-Mass Spectrometry) ESI system/HRESIMS (High Resolution-Electrospray Ionisation Mass Spectroscopy). The crude extract obtained was 0.4023 g and the yield was 0.33% (w/w). Fraction V from KLTP separation was known to have antibacterial activity against E.coli. The inhibition shown by the fraction V was only 1.5 mm at a concentration of 1000 ppm. LC-MS/HRESIMS analysis of the fraction V indicated that it contained 9-octadecenamide.
