VALIDASI METODE ANALISIS UNTUK PENENTUAN TOTAL MERKURI DALAM DAGING IKAN MENGGUNAKAN PENCERNAAN MIKROGELOMBANG SECARA SPEKTROFOTOMETRI SERAPAN ATOM UAP DINGIN; VALIDATION OF ANALYTICAL METHOD FOR DETERMINATION TOTAL MERCURY IN FISH MEAT USING MICROWAVE DIGESTION BY COLD VAPOUR ATOMIC ABSORPTION SPECTROPHOTOMETRY

2015 | Skripsi | FMIPAIn this research has done validation analytical method for determination total mercury in fish meat using microwave digestion. This method is one of application microwave for determination total mercury in fish meat sample. Besides that, this research aims to study the effect of sample destruction using close system and open system by cold vapour atomic absorption spectrofotometry. This research studies the stability of sample destruction using close system by microwave digestion and it compares with open system by hotplate. The stability has been assested by reproducibility and repeatibility. Validation test using determination of linearity, method detection limit (MDL), limit of quantification (LOQ), precision, accuration, % trueness, % bias. A statistical t-test has been applied for comparing the analysis result by microwave digestion and hotplate if the analysis by both methods are significantly different or not. The result showed that destruction using microwave digestion is more rapidly and more stable than using hotplate, it showed by %RSD microwave digestion < %RSD hotplate, 3,293% < 6,305%. The parameters validation using CRM DORM-4 showed that analytical method for determination total mercury in fish meat using microwave digestion are good, it showed by linearity 0,999, Limit of Linearity (LOL) 10 ng/mL, Limit of Quantification (LOQ) 0,468 ng/mL, Method Detection Limit (MDL) 0,146 ng/mL, accuration 100,243%, % trueness 100,243%, % bias 0,243%. The result of t-test showed that the analysis result by both microwave digestion and hotplate were not significantly different. It can be concluded that analytical method for determination total mercury in fish meat using microwave digestion can be used in the laboratory.
