VALIDASI METODE ANALISIS NaDBS DALAM IKAN LELE SECARA MASERASI DAN SPEKTROFOTOMETRI UV-VIS MENGGUNAKAN MALASIT HIJAU; METHOD VALIDATION OF NaDBS ANALYSIS IN CATFISH BY MACERATION AND UV-VIS SPECTROPHOTOMETRY USING MALACHITE GREEN

This research aimed to validate the extraction method of NaDBS by maceration and the analysis by UVVis spectrophotometry in cultivated catfish, study the relationship between NaDBS exposures in cultivated water with NaDBS concentrations accumulated in catfish, and determine of NaDBS contents in consumption fish, such as: catfish, tilapia and carp. This research began with method validation of NaDBS analysis in cultivated water and catfish. NaDBS extraction was performed by maceration using methanol solvent for 16 h. The analysis of NaDBS was performed by UV-Vis spectrophotometry with malachite green. Then, analysis method of NaDBS applied in consumption fish. The method validation parameters of NaDBS analysis in cultivated water provided linearity 0.9964, LOD 0.03090 mg/L, LOQ 0.09360 mg/L, sensitivity 12.29 × 104 L mol-1 cm-1, precision < 2%, and accuracy 98.35-99.88%. Meanwhile, the method validation parameters of NaDBS analysis in cultivated catfish provided linearity 0.9970, LOD 0.01910 mg/L, LOQ 0.05790 mg/L, sensitivity 14.45 × 104 L mol-1 cm-1, precision < 2%, and accuracy 87.74-90.02%. NaDBS concentrations in catfish that farmed in water containing NaDBS increased with increasing of NaDBS contents in cultivated water. NaDBS in catfish showed the accumulation in their bodies. BCF values were inversely proportional to the contents of NaDBS in cultivated water, each by 1.199; 0.7015; 0.4336 and 0.3377 L/kg. Consumption fish samples (catfish, tilapia, and carp) were found to contain NaDBS 2.991-8.132 mg/kg. These results showed that analysis method of NaDBS by maceration and UV-Vis spectrophotometry using malachite green was very good and could be applied in fish. The analysis results in consumption fish showed the bad condition if consumed by humans. Therefore, the analysis method of NaDBS could also be used in other species of fish.
