MODEL MATEMATIKA REGULASI PERBAIKAN SEL AKIBAT AKTIVASI VIRUS EPSTEIN-BARR SEBAGAI INDIKASI AWAL PENYAKIT KARSINOMA NASOFARING; A MATHEMATICAL MODEL FOR CELL REPAIR REGULATION AFTER EPSTEIN-BARR VIRUS ACTIVATION AS EARLY INDICATION OF NASOPHARYNGEAL CARCINOMA DISEASE

2015 | Skripsi | FMIPA UGMNasopharyngeal Carcinoma (NPC) is a tumor which grows on nasopharynx epithelial cell. This cancer is caused by the activation of Epstein-Barr Virus (EBV) in the body triggered by environmental factor. In this thesis, the writer constructs and simulates a Mathematical model for cell repair regulation which occurs in the body to respond DNA damage caused by EBV activation. The purpose of the simulation is to understand characteristic of NPC disease through fluctuation of proteins expression in this regulation during EBV host cell proliferation phase and after mutation on p53 gene happens as early indication of NPC tumor growth. Another purpose of the simulation is to understand therapy effect by using Nutlin-3a molecule in order to prevent (to delay) the occurrence of mutation on p53 gene, so that NPC disease does not occurs. During EBV host cell proliferation phase, phosphorylation and acetylation of inactive p53 protein always happen to respond DNA damage. After p53 gene mutation happened, expressions of phosphorylated and acetylated p53 protein decrease suddenly, because the proteins undergo inactivation. Then, expression of proteins involved in cell repair regulation oscillate continuously as a sign of chromosomal instability phenomena on their gene which causes NPC tumor growth. Early therapy using Nutlin-3a molecule on patient who has accumulation of DNA damage in his/her nasopharynx epithelial cell is effective to reduce the accumulation of DNA damage, so that phosphorylated and acetylated p53 protein expressions remain stable. It describes that mutation on p53 gene as the cause of NPC tumor growth can be prevented (delayed), so early therapy using Nutlin-3a molecule can become medical recommendation to prevent (to delay) NPC tumor growth.
