ISOLASI METABOLIT SEKUNDER DARI DAUN BANGUN-BANGUN (Coleus amboinicus Lour.) SERTA POTENSINYA SEBAGAI ANTIOKSIDAN; ISOLATION OF SECONDARY METABOLITES FROM BANGUN BANGUN LEAVES (Coleus amboinicus Lour.) AS WELL AS POTENTIAL ANTIOXIDANT

The First, samples of leaves bangun-bangun were prepared and dried in the open space by avoiding direct exposure to sunlight, and the sample size by using a blender. The Second, maceration process using methanol solvent. The third, the extract was filtered, concentrated by an evaporator, a partition based on the increase in the polarity of solvent (n-hexane, chlorofom, and ethyl acetate), phytochemical test, and preliminary testing of each antioxidant extracts with DPPH. The fourth, extracts with higher activity was purified by column chromatography. The fifth, active isolates (crystal) were analyzed for melting point, UV, IR, GC-MS, 1H-NMR and 13C-NMR. Isolates obtained results column two crystals with a melting point of 114-115 °C and 232-233 °C, crystal was obtained dissolve completely with methanol, ethyl acetate and chloroform. The results of GC-MS analysis of the active isolates I, there is a retention time peak with 7.316 minutes and percent peak 100%. Active isolates II, there are two peaks, the first retention time of 6.658 minutes with percent peak 3.95%, the socond has retention time of9.001 minutes with percent peak 96.05%. Each crystal has antioxidant activity (IC 50) of 388.40 and 338.15 including antioxidant active compounds into the weak category. The first active isolates were 7a-acetoxyroyleanone-16-hidroxy, and isolates the active second were 7a-hidroxyroyleanone-16-acetoxy.
