KARAKTERISASI FRAGMEN 0,58 kb GEN PENGKODE STILBEN SINTASE (STS) DARI TANAMAN MELINJO (Gnetum gnemon L.); CHARACTERIZATION OF 0.58 kb GENE FRAGMENT ENCODING STILBENE SYNTHASE FROM MELINJO PLANT (Gnetum gnemon L.)

A study on the characterization of 0.58 kb gene fragment encoding stilbene synthase from melinjo (Gnetum gnemon L.) has been done. The aim of this study is to obtain the sequences of sts gene fragment from melinjo. Amplification of the gene fragment by RT-PCR (Reverse Transcriptase Polymerase Chain Reaction) was started by primers designing based of conserved sequence amino acid from 10 STS which have been published in genebank. The results of primer design was synthesized and used for RT-PCR amplification using two step technique using total RNA isolated from melinjo’s leaves as template. The results of RT-PCR fragment was then isolated, purified and sequenced. The sequences were analyzed by comparing the nucleotide sequence of existing sts genes in genebank. The designed primer was named as GGF2 (5' GTTCCACCTGCGAAGCAGCC 3') pair with GGR2 (5' CTGGATCGCACATCCTGGTG 3') which is predicted in position 334-354 and 897-916. RT-PCR amplification result in several DNA fragments with one of them has size 0.58 kb which is expected result. Nucleotide Sequence of isolated 0.58 kb fragment shows 67% insimilarity to Arachis hypogaea L00952 sts gene. This homology is higher than the average similarity among sts genes which is 66%. Thus, the amplified fragment is a part of sts gene from melinjo.
