2013 | Disertasi | PROGRAM STUDI S2 ILMU KIMIAThe effect of deacetylation degree of chitosan on the effectiveness of chitosan beads as solid support immobilized lipase by cross-linking techique has been carried out. The study aims to obtain optimal conditions for immobilization of lipase on chitosan beads. Chitosan beads made from the chitosan powder with various degree of deacetylation by inverse phase method. The study starts from the optimization process of immobilization such as phosphate buffer pH and concentration of glutaraldehyde as a cross-linker. Lipase immobilized on chitosan beads was tested activities, thermal stability, re-uses and parameter kinetics on the hydrolysis reaction of palm oil triglycerides. Phospate buffer with pH 6 is optimal condition to dissolved lipase enzym. DD chitosan that resulted are 80,71%, 87,51% and 91,77%. Optimum concentration of glutaraldehyde optimal is approached in glutaraldehyde 0,5%(v/v) for DD chitosan bead 80,71% with (%) percentage immobilization enzyme 84% ; 1,5%(v/v) for DD chitosan bead 87,51% with (%) percentage immobilization enzyme 91% and 1,5%(v/v) for DD chitosan bead 91,77% with (%) percentage immobilization enzyme 82%. Specified activities immobilized of lipase enzymes approached of chitosan bead %DD 80,71; %DD 87,51 and %DD 91,77 are 82, 85 and 81 U/g, but specified activity of free lipase enzyme is 145 U/g. Substrate activity immobilized enzyme of chitosan bead %DD 80,71; %DD 87,51 dan %DD 91,77 pointed from KM 0,116; 0,101; 0,122. Lipase immobilized have thermal stabilized and stabilized re-used is better than free lipase enzyme. Looked from DD chitosan bead that used in immobilize lipase enzyme, DD chitosan bead 91,77% has the best thermal stabilized and stabilized re-use.
