EFEK JUS BATANG BROTOWALI (Tinospora crispa L. Miers) TERHADAP GAMBARAN LEUKOSIT TIKUS WISTAR YANG DIINDUKSI ALOKSAN

Diabetes melitus adalah penyakit metabolik yang ditandai dengan hiperglikemia. Obat sintetis yang digunakan saat ini tidak cukup efektif, selain memiliki efek samping, obat tersebut juga memerlukan biaya cukup mahal. Salah satu obat alternatif yang digunakan untuk mengatasi penyakit diabetes melitus adalah tanaman Brotowali (Tinospora crispa L. Miers). Penelitian ini bertujuan untuk mengetahui peranan jus batang Brotowali terhadap gambaran leukosit (total leukosit dan diferensial leukosit) tikus penderita diabetes. Tikus putih betina galur Wistar sebanyak 25 ekor umur 3 bulan yang terbagi dalam 5 kelompok dan masing-masing kelompok terdiri dari 5 ekor.Tikus kelompok I, II, III dan IV diinjeksi 3 kali Aloksan tetrahidrat dengan dosis 150 mg/kg BB/ekor secara intraperitoneal yang dilarutkan dalam larutan HCL 0,01M sedangkan kelompok V tidak diinjeksi Aloksan sebagai kontrol negatif.Selanjutnya tikus kelompok I diobati jus batang Brotowali 18 mg/200 g BB/ekor/hari secara peroral. Tikus kelompok II diobati jus batang Brotowali 9 mg/200 g BB/ekor/hari secara peroral. Tikus kelompok III diobati ekstrak batang Brotowali 9 mg/200 g BB/ekor/hari/PO (dibunuh). Tikus kelompok IV tanpa diobati jus batang Brotowali digunakan sebagai kontrol positif. Tikus Kelompok V tanpa injeksi Aloksan namun diobati jus batang Brotowali 9 mg/200 g BB/ekor/hari/PO digunakan sebagai kontrol negatif. Pemberian Jus batang Brotowali dilakukan selama 22 hari dimulai pada hari ke-7. Pengambilan darah dilakukan sebelum tikus diinjeksi Aloksan (hari ke-0), setelah tikus diberi jus batang Brotowali (hari ke-16) dan pada akhir penelitian (hari ke-28). Parameter yang diukur adalah gambaran leukosit(total leukosit dan diferensial leukosit).Selama penelitian diikuti perubahan gejala klinis yang terjadi. Data gambaran leukosit(total leukosit dan diferensial leukosit) diolah secara statistik dengan menggunakan ANOVAOne Way(Analysis of Variance) pada taraf signifikansi 5% danuji lanjutan Least Significantly Difference (LSD) Hasil penelitian menunjukkan bahwa induksi diabetes dengan Aloksan tetrahidrat mengakibatkan kenaikan jumlah total leukosit dan penurunan jumlah sel limfosit, pemberian jus batang Brotowalidosis 9 mg/200 gBB dan dosis 18 mg/200gBB dapat mengembalikan gambaran leukosit(total leukosit dan diferensial leukosit)menjadi normal.

Diabetes mellitus is a metabolic disease characterized by hyperglycemia. Synthetic drugs are not effective enough, it has side effect and need a cost quite expensive. One of the alternative medicines used to treat diabetes mellitus is a Brotowali (Tinospora crispa L. Miers). The aim of this research is to understand the effect of Brotowali stem juice to leukocyte profile (total leukocyte count and differential leukocyte) of diabetic rats Twenty five rats of wistar strain age three months were divided into five group, each group consist of five rats. group I,II,III,IV were injected by Alloxan tetrahydrat three times at a dose of 150mg/kg BW/IP were dissolved 0,01 M HCL solution and group V without injection of Alloxan was used as a negative control. Group I treated Brotowali stem juice 18mg/200 g Bw /day orally. Group II treated Brotowali stem juice 9 mg/200 g BW/day orally.Group III treated Brotowali stem juice 18mg/200 g BW/day orally (Killed). Group IV without treated Brotowali stem juice as positive control. Group V without injected Alloxan but treated Brotowali stem juice 9mg/200 g BW/day orally as negative control. The treatments were given once daily for 22 days and start at seventh day of study. Blood sampling was done before alloxan injection (day 0), after administration of Brotowali stem juice (day 16), and at the end of research (day 28). Parameters measured were leukocytes ( leukocyte total count and differential leukocyte ) .Clinical symptoms were observed during the study. The result of leukocyte was analyzed by One Way ANOVA at significance level of 5% andLeast Significantly Difference (LSD). Parameters measured were leukocyte (total leukocyte count and differential leukocyte ). The result of this research showed Aloxan induced diabetes made increase leukocyte total count and decrease number of lymphocite. Brotowali stem juice dose 9 mg/ 200 gBW and 18 mg/ 200 gBW can restore the leukocyte (total leukocyte count and differential leukocyte ) of diabetic rats.
