PENGARUH SUHU THAWING TERHADAP KUALITAS MOTILITAS SEMEN BEKU POST THAWING SAPI POTONG SIMMENTAL

2014 | Skripsi |Penelitian ini bertujuan untuk mengetahui pengaruh motilitas spermatozoa pada semen beku Sapi Simmental post thawing terhadap media bersuhu air keran, air bersuhu 35˚C - 37˚C, dan air es dalam waktu inkubasi < 1 menit, 10 menit, 30 menit, dan 5 jam. Penelitian ini dilakukan dengan menggunakan 30 straw semen beku sapi Simmental yang diperoleh dari Balai inseminasi buatan Ungaran. Sepuluh straw sapi Simmental yang berisi semen beku dithawing dengan media air keran, air hangat bersuhu 35˚C - 37˚C, dan air es secara berurutan. Kemudian, diinkubasikan kedalam incubator bersuhu 38.5˚C dalam rentang waktu < 1 menit, 10 menit, 30 menit dan 5 jam. Pemeriksaan motilitas individu spermatozoa dilakukan dengan metode pengamatan pasca tiap waktu inkubasi dengan menggunakan mikroskop. Hasil penelitian ini dapat disimpulkan bahwa thawing semen beku dapat dilakukan dengan menggunakan air keran, air hangat bersuhu 35 ˚C -37˚C, maupun air es, namun terlihat kecenderungan persentase motilitas spermatozoa pasca thawing yang lebih tinggi ketika semen beku dithawing dengan menggunakan air hangat bersuhu 35 ˚C -37˚C. Pada semua perlakuan, presentase motilitas menurun secara signifikan seiring bertambahnya waktu inkubasi (P < 0,05)
This research aims to understand the influence of sperm motility of post - thawed Simmental bull semen in various temperature medium; tap-water temperature, heated water (35˚C - 37˚C) , and iced-water temperature then incubated in less than 1 minute, 10 minutes, 30 minutes, and 5 hours period. This research performed 30 straws frozen semen of Simmental bull which is obtained from BIB Ungaran. Every ten straws which is filled with frozen Simmental bull semen were thawed with tap water, heated water (35˚C - 37˚C), and iced-water sequentially. It was then being incubated in the incubator with temperature of 38.5˚C in a span of less than 1 minute, 10 minutes, 30 minutes and 5 hours. The examination of individual motility of spermatozoa was performed by the observation method using a microscope after each incubation time. As results, thawing on the frozen semen could be done using tap-water, heated water (35˚C - 37˚C), as well as iced water. However, the trend of percentage of sperm motility after thawing is higher when it is done using heated water in 35˚C - 37˚C temperature. The percentage of motility declined significantly with the increase of incubation time in every treatment (P < 0,05)
