PENGARUH OVARIEKTOMI TERHADAP RASIO BERAT HATI DAN GAMBARAN HISTOPATOLOGI HATI TIKUS Sprague Dawley

2014 | Skripsi |Penelitian ini dilakukan untuk mengetahui pengaruh ovariektomi terhadap berat badan, rasio berat hati terhadap berat badan dan gambaran histopatologi hati tikus Sprague Dawley. Delapan tikus Sprague Dawley betina umur 2 bulan, dibagi 2 kelompok masing-masing 4 tikus. Tikus kelompok A dioperasi semu (non ovariektomi), tikus kelompok B dioperasi ovariektomi. Sebelum diovariektomi, tikus dianastesi dengan Ketamin 10 % (50 mg/kg) dan Xylazin 2% (5 mg/kg) . Setelah dua bulan pasca operasi ovariektomi, tikus dinekropsi untuk pengambilan hati. Hati ditimbang, kemudian dimasukkan dalam larutan NBF 10 % (Neutral Buffered Formalin) untuk dibuat preparat histopatologi dengan pengecatan HE (Hematoxylin Eosin). Hasil penelitian menunjukkan berat badan tikus ovariektomi (203,5 + 12,01 gram) secara bermakna lebih tinggi dibandingkan tikus non ovariektomi (173,3 + 21,3 gram) (P<0,05). Rasio berat hati terhadap berat badan tikus non ovariektomi (0.033 + 0.0035) tidak berbeda bermakna tikus ovariektomi (0.032 + 0.0009) (P>0,05). Gambaran makroskopik hati tikus ovariektomi menunjukkan tidak ada perubahan, sedangkan gambaran histopatologi hati tikus ovariektomi menunjukkan adanya perlemakan. Kesimpulan, dua bulan pasca tikus Spargue Dawley diovariektomi,tikus mengalami peningkatan berat badan, meskipun rasio berat hati terhadap berat badan tidak berbeda bermakna. Gambaran makroskopik hati tikus ovariektomi tidak ada perubahan, sedangkan gambaran histopatologi hati tikus ovariektomi menunjukkan adanya perlemakan.
This research was conducted to study the effect ovariectomy on liver weight ratio and histopatology of Spargue Dawley rat liver two months after ovariectomy. Eight femele Sprague-Dawley rats at 2 months of age were,divided into 2 groups of 4 mice each. Group A was sham operated (non ovariectomy) and group B was ovariectomy. Ketamine (50 mg / kg) and Xylazin 2% (5 mg / kg) were use as an anasthetic agent. Two months after ovariectomy the rat was necropsized to remove the liver. Liver were taken and weighed first, then put it in 10 % formalin solution for histopathological examination using Hematoxyline Eosin staning. The results showed that the body weight ovariectomized rat (203.5 + 12.01 grams) was significantly higher than rats non-ovariectomized (173.3 + 21.3 g) (P <0.05). Ratio of livet weight to body weight of ovariectomized (0.032 + 0.0009) was not significantly higher than non-ovariectomized (0.033 + 0.0035) (P> 0.05). Macroscopic figure of the liver of ovariectomized was not shown any change where as non-ovariectomized. However there was fatty histopatologically. Based on the result, two months after Spargue Dawley rats ovariectomized had increased body weight, although rat non-ovariectomized weight did not different significantly. Macroscopic figure of the liver of rats not shown any change, while the histopathological figure of the liver showed fatty of ovariectomized rats.
