STUDI HISTOLOGI DUODENUM LOWO (Myotis sp.)

2014 | Skripsi |Lowo (Myotis sp.) merupakan satwa liar yang termasuk kelas Mamalia, ordo Chiroptera dan famili Vespertilionidae. Lowo merupakan salah satu kelelawar dari subordo Microchiroptera yang umumnya memakan serangga. Duodenum memiliki fungsi utama sebagai pelanjut proses digesti pada intestinum tenue dan absorpsi nutrisi dari makanan yang dimakan. Perbedaan kebiasaan pakan lowo dengan mamalia lainnya ini menyebabkan perlunya dilakukan penelitian mengenai histologi duodenum lowo. Penelitian ini menggunakan lima duodenum lowo berasal dari Magelang. Jaringan diproses dengan metode parafin, kemudian jaringan dipotong dengan ketebalan 5-6μm. Jaringan dalam coated slide diwarnai dengan Hematoksilin Eosin (HE) dan pewarnaan Alcian Blue pH 2,5-Periodic Acid Schiff (AB pH 2,5-PAS). Hasil dianalisis secara deskriptif. Lowo hasil penelitian dapat disimpulkan struktur histologi duodenum tersusun atas tunika mukosa, tunika submukosa, tunika muskularis dan tunika serosa. Kelenjar Brunner ditemukan pada bagian diantara daerah akhir sfringter pilori lambung sampai bagian awal duodenum dan bersifat serous. Panjang kelenjar Brunner dengan rata-rata 686,5 ± 89,62 μm. Hasil pewarnaan AB pH 2,5 PAS menunjukkan sel goblet pada lamina epitelialis mukosae dan pada kripte lieberkuhn positif pewarnaan Alcian Blue pH 2,5, sedangkan sel absorpsi, sel kolumner pada kripte lieberkuhn dan kelenjar Brunner positif pewarnaan Periodic Acid Schiff. Distribusi mukopolisakarida asam ditemukan pada sel goblet di lamina epitelialis mukosae dan sel goblet di kripte lieberkuhn, sedangkan distribusi mukopolisakarida netral ditemukan di sel absorpsi, sel kolumner pada kripte lieberkuhn dan kelenjar Brunner pada tunika submukosa.
Lowo (Myotis sp.) is one at Indonesia wild animals that belongs to the class Mammalians, order Chiroptera, suborder Microchiroptera and family Vespertilionidae. Thus, it is one of the bats from suborder Microchiroptera that feed on insect. The duodenum has two main function, to digest food as a part of the small intestine, and absorb nutrition from the digested food. This unique feeding behavior of lowo is the reason behind this research about the histology of lowo’s duodenum. In this study, five lowo’s duodenum from Magelang were collected. Tissues were processed with parafin method, then cut in 5-6m thickness and mounted in coated slide. Tissues inside the coated slide are then stained using the Hematoxylin Eosine (HE) and Alcian Blue pH 2,5-Periodic Acid Schiff (AB pH 2,5-PAS). The tissue were examide microscopically and is analyzed descriptively. Based on the observation, it can be concluded that the histological structure of lowo’s duodenum is composed of tunica mucosa, tunica submucosa, tunica muscularis, and tunica serosa. Brunner glands’ characteristic are serous, distributed from the end of pyloric sphingter to the beginning of the duodenum. The average length of each gland is 686,5 + 89,62 m. Goblet cells are distributed in the lamina epithelial mucosae and lieberkuhn crypt, and they are react positively to the Alcian blue pH 2,5 staining, while the absorbtion cells, columnar cells in the lieberkuhn crypt, and Brunner glands react positively to Periodic Acid Schiff staining. The distribution of acid mucopolysaccharides are found in the goblet cells in lamina epithelial mucosae and goblet cells of lieberkuhn crypt. Neutral muccopolysaccharides are found in the absorbtion cells, columnar cells of lieberkuhn crypt, and Brunner glands in tunica submucosa.
