EFEK PENAMBAHAN EKSTRAK TEH HIJAU PADA PENGENCER TRIS-KUNING TELUR TERHADAP KUALITAS SPERMATOZOA SEGAR EPIDIDIMIS SAPI

Tujuan dari penelitian ini untuk mengetahui efek penambahan ekstrak teh hijau dengan konsentrasi 0,05%, 0,1% dan 0,15% pada pengencer Tris-kuning telur sebagai antioksidan untuk meningkatkan kualitas spermatozoa epididimis sapi. Metode koleksi spermatozoa dilakukan dengan mencacah epididimis sapi. Pengencer Tris-Kuning telur dengan konsentrasi ekstrak teh hijau yang berbeda ditambahkan ke dalam spermatozoa. Evaluasi motilitas dan prosentase hidup dilakukan secara mikroskopik. Hasil dari penelitian ini menunjukkan bahwa motilitas dan prosentase hidup pada pengencer ekstrak teh hijau 0,05% , 0,1% dan 0,15% secara berturut-turut (7,50%; 27,25%), (12,50%; 31,75%) dan (20,00%; 41,00%) memiliki kualitas yang lebih tinggi dibandingkan dengan motilitas dan prosentase hidup kontrol (5,00%; 26,50). Data absolut menunjukkan kecenderungan kenaikan motilitas dan prosentase hidup spermatozoa. Kesimpulan dari penelitian ini yaitu pada penambahan ekstrak teh hijau konsentrasi 0,15% memberikan tingkat kualitas spermatozoa tertinggi. The objective of this study was to determined the effect of green tea extract with 0,05%, 0,1% and 0,15% concentration that added on Tris-egg yolk dilution as antioxidant to increase of spermatozoa quality, which collected from bull epididymis. Spermatozoa collection method have done by slicing bull epididymis. Tris-egg yolk dilution with different concentration of green tea extract added into the spermatozoa. Evaluation of motility and viability have done microscopically. That results showed that motility and viability percentation on green tea extract dilution 0,05% , 0,1% and 0,15%, were (7,50%; 27,25%), (12,50%; 31,75%) and (20,00%; 41,00) respectively, had a higher motility and viability percentation compare to the control (5,00%; 26,50%). The data showed this tend to the increase motility and percentage of live spermatozoa. The conclusion of this study define that the addition of green tea extract 0,15% had the highest sperm quality improvement. Key Word : Green Tea Extract, Bull Epididymis, Spermatozoa
