Gambaran Mikroskopis Uterus Tikus Wistar Ovariektomi yang diberi Suplementasi Vitamin D3

2012 | Skripsi |Penelitian ini dilakukan untuk mengkaji suplementasi kalsitriol pada tikus Wistar ovariektomi terhadap gambaran histopatologis uterus. Sepuluh tikus Wistar ovariektomi umur 9 minggu, secara acak dibagi menjadi 2 kelompok masing-masing 5 tikus. Tikus kelompok ovariektomi yang diberi pakan standar (grup I) dan tikus kelompok ovariektomi yang diberi pakan standar dan kalsitriol 8 ng (grup II). Pada umur 15 minggu tikus dieuthanasi menggunakan anastetika ketamin 10% dosis 50 mg/kg berat badan dan xylasin 2% dosis 5 mg/kg berat badan. Uterus diambil dan difiksasi dalam formalin 10% untuk pemeriksaan mikroskopis. Hasil penelitian gambaran histopatologis uterus pada tikus Wistar yang diovariektomi rata-rata panjang epitel adalah 17,52±5,69 μm, ketebalan endometrium adalah 462,47±203,08 μm, dan jumlah kelenjar adalah 22,6±9,4. Gambaran histopatologis uterus tikus Wistar ovariektomi yang diberi suplemen kalsitriol ratarata panjang epitel adalah 13,68±4,81 μm, ketebalan endometrium 368,40±192,96 μm, dan jumlah kelenjar adalah 18,8±12,2. Analisis statistik menunjukkan bahwa tidak ada perbedaan yang signifikan antara panjang epitel lamina epitelial mukosa endometrium uterus dan jumlah kelenjar pada grup I dan grup II. Namun, ada perbedaan signifikan pada ketebalan endometrium uterus antara grup I dan grup II. Kesimpulan, suplemen kalsitriol menyebabkan penurunan ketebalan endometrium, atropi (pemendekan) epitel lamina epithelial mukosa, dan penurunan jumlah kelenjar pada endometrium uterus tikus Wistar ovariektomi. The objective of the study was to evaluate calcitriol suplementation on histophatological feature of ovariectomized uterine in Wistar rat. Ten female Wistar rats, at 9 weeks of age were randomly divided into two group of five. Ovariectomized group rats were fed standard diet (group I) and ovariectomized group rats was fed standard diet plus 8 ng of calcitriol suplementation (group II). At 15 weeks of age, rats were euthanized with 50 mg/kg BB of 10% ketamine and 5 mg/kg BB of 2% xylasin, the uterine was removed and was immersed in 10% formalin for microscopic examination. Result of study show that ovariectomized uterine of Wistar rat, have length of the epithelium was 17,52±5,69 μm, the thickness of the endometrium was 462,47±203,08 μm, and number of uterine glands was 22,6±9,4. The histopathological feature of ovariectomized uterine of Wistar rats plus calcitriol suplementation, show length of the epithelium was 13,68±4,81 μm, the thickness of the endometrium was 368,40±192,96 μm, and number of uterine glands was 18,8±12,2. Statistical analysis show that there is not significant difference between the number of uterine glands and the length of the epitelium in group I and group II. However there is significant difference in the thickness of endometrium between group I and group II. Conclusion, calcitriol supplementation decreased the thickness of endometrium, atrophy (shortening) in epithelium lamina epithelial mucosa, and decreased the number of uterine glands in endometrium uterine of ovariectomized Wistar rats.
