Sebanyak20sampelsususapidikulturpadamediaPlatAgarDarah(PAD)danMacConkeyAgar(MCA)kemudiandiinkubasipadasuhu37ºCselama24jam.Koloniyangtumbuhdikelompokkansesuaidenganmorfologikoloninyadandilanjutkanuji-ujibiokimiadenganmenggunakanujigula-gula,SulphideIndolMotility(SIM),Indol,TryptitaseSoyAgar(TSA)danSimonCitrate.UntukmengetahuisensitivitasterhadapantibiotikaKanamycindanNovobiocinbakteriyangtelahdikulturpadamediaBrainHeartInfussion(BHI)kemudiandikulturpadamediaMullerHintonAgar(MHA)dandiukurzonahambatsetelahdiinkubasipadasuhu37ºCselama24jam.H asilpenelitianmenunjukkanbahwadari20sampelsususapidiperoleh11sampelterinfeksibakteriEscherichiacoli.Ujisensitivitasmenunjukkanbahwa7isolat(63,6%)sensitifterhadapkanamycin,4isolat(36,3%)intermedietterhadapkanamycin,sedangkanuntuknovobiocin10isolat(90,9%)resistenterhadapnovobiocindan1isolat(9,09%)sensitifterhadapnovobiocin.K atakunci:Escherichiacoli,mastitis,subklinis,kanamycin,novobiocin
Mastitisisoneofmanydiseasesthatcancauseanyharm.Thisresearchisaimedtoidentifyisolation,identificationandthesensitivitytestofEscherichiacolicauseofsubclinicalmastitisagainstkanamycinandnovobiocinantibiotics. Twentysamplesofcow’smilkareculturedonBloodplateagar(BPA)andMacConkeyAgar(MCA)andincubatedat37oCfor24hours.Coloniesthatgrowaregroupedaccordingtotheircolonymorphologyandthencontinuedbybiochemicaltestwhichareconfectionerytest,SulphideIndolMotility(SIM),Indol,TryptitaseSoyAgar(TSA)danSimonCitrate.ToknowthesensitivityofbacteriatoKanamycinandNovobiocinantibiotics,thebacteriathathavebeenculturedonBrainHeartInfussion(BHI),andthenculturedagainonMullerHintonAgar(MHA)aremeasuredofresistancezoneafterincubatedfor24hourson37oC. Theresultsshowedthatonceconfirmed,fromtwentysamplesofcow’smilk,obtainelevensamplesthatinfectedofEscherichiacolibacteria.Thesensitivitytestshowedthatsevenisolate(63,6%)aresensitivetoKanamycin,fourisolate(36,3%)areintermediate-sensitivetoKanamycin.WhileforNovobiocin,tenisolate(90,9%)areresistanceagainstit,andoneisolate(9,09%)aresensitiveofit.
