Pengaruh Suplementasi Kalsitriol Terhadap Gambaran Histopatologis Uterus Tikus Wistar Pasca Ovariektomi

2012 | Skripsi |Penelitian ini bertujuan untuk mengetahui pengaruh suplementasi kalsitriol selama 1,5 bulan terhadap gambaran histopatologis uterus tikus Wistar pasca ovariektomi. Lima belas ekor tikus Wistar betina umur 8 minggu, secara acak dibagi tiga kelompok (kontrol/K, ovariektomi/OV dan ovariektomi+kalsitriol/ OVD) masing-masing 5 tikus. Tikus kelompok K dan OV diberi pakan standar, sedangkan tikus kelompok OVD diberi pakan standar+kalsitriol 8 ng. Pada umur 15 minggu, tikus dieutanasi. Uterus diambil untuk pemeriksaan histopatologis dengan pewarnaan Hematoxylin-Eosin. Hasil pemeriksaan histopatologis uterus pada panjang epitel lamina mukosa dan jumlah kelenjar endometrium uterus antara kelompok tikus K, OV dan OVD tidak berbeda signifikan, namun pada tebal endometrium uterus antara kelompok tikus K, OV dan OVD berbeda signifikan. Uji LSD memperlihatkan tebal endometrium pada tikus OVD dan tikus OV tidak berbeda signifikan, tikus OVD berbeda signifikan dengan tikus K, tikus OV berbeda signifikan dengan tikus K (p<0,05). Hasil penelitian dapat disimpulkan bahwa ovariektomi secara bermakna dapat menyebabkan atropi uterus yang ditandai dengan penurunan ketebalan endometrium. Suplementasi kalsitriol tidak berpengaruh terhadap penurunan panjang epitel, ketebalan endometrium, jumlah kelenjar endometrium uterus pada tikus ovariektomi. This study was conducted to determine the effect of calcitriol supplementation for 1.5 months on histopathological feature of ovariectomized uterine in Wistar rat. Fifteen female Wistar rats at 8 weeks of aged, were randomly divided into three groups (control group/K, ovariectomized group/OV, and ovariectomized group+calcitriol/OVD) in wich group consisted of 5 rats. Group K and OV rats were fed standard diet, while group OVD rats was fed standard diet+8 ng of calcitriol. At 15 weeks of age, the rats were euthanized. The uterine was removed for histopathological examination with Hematoxylin-Eosin staining. The results of the research showed that there was no significant difference among group K, OV, and OVD in case of length of epithelium and the number of endometrial glands. However, there was significant difference in the endometrial thickness among group K, OV, and OVD. Statistical analysis using LSD test showed that there was no significant difference among group OVD and OV in the endometrial thickness, whereas if both treatments compared with group K showed significant difference (p<0.05). To conclude, ovariectomy result in decreased of the uterine endometrial thickness. Whereas, calcitriol supplementation did not affect the length of uterine epithelium, endometrial thickness, the number of uterine glands in ovariectomized rats.
