Purwoceng Gunung sebagai aprodisiaka mengandung komponen kimia kelompok steroid, atsiri, furanokumarin, dan vitamin, yang terdapat di bagian akar. Kelompok steroid terdiri dari sitosterol, stigmasterol (stigmasta-7, 16 dien-3-ol), dan (stigmasta-7, 25 dien-3-ol). Komponen kimia tersebut yang menjadikan Purwoceng Gunung sebagai obat tradisional untuk meningkatkan vitalitas dan kesuburan pria. Stigmasterol memiliki kelarutan yang sangat rendah dalam air, karena memiliki sifat semi polar hingga non polar. Penelitian ini bertujuan untuk mengetahui formula optimal S-SNEDDS ekstrak akar Purwoceng Gunung dengan minyak kemiri sebagai fase minyak dan mengetahui peningkatan persentase disolusi stigmasterol secara in vitro. Ekstrak akar Purwoceng Gunung dibuat dengan metode maserasi menggunakan etanol 96%, selanjutnya dengan n-heksan:kloroform (1:1). Ekstrak yang dihasilkan dianalisa kandungan stigmasterolnya dengan HPLC. Formula SNEDDS yang digunakan adalah minyak kemiri 15,00 % b/b, Tween 80 55,76% b/b, Croduret® 9,84 %b/b, dan propilen glikol 19,40 %b/b. Maltodextrin digunakan sebagai solidifying agent menggunakan metode spray drying. S-SNEDDS dikarakterisasi ukuran tetesan dan distribusi partikelnya, sudut diam (angle of repose), bulk density, tapped density, drug content, morfologi serbuk, stabilitas, emulsification time, dan uji disolusi in vitro serta kinetika pelepasannya. Ekstrak akar Purwoceng Gunung dengan rendemen 9,61% mengandung stigmasterol dengan kadar 11,96 + 0,93 %b/b, tekstur kental, berwarna coklat hijau kehitaman, rasa pahit dan bau khas Purwoceng, kadar air 3,53 + 0,54, dan kandungan logam berat Pb (<0,03 mg/kg), Cd (<0,01 mg/kg), dan Hg (22,93 µg/kg). Hasil uji kadar air dan kandungan logam berat memenuhi persyaratan yang ditetapkan BPOM RI. Extract loading dalam SNEDDS 150 mg/g dengan rata-rata ukuran partikel 21,50 nm. S-SNEDDS mengandung stigmasterol dengan kadar 4,14 + 0,86 %b/b, memiliki sifat alir yang buruk, waktu emulsifikasi 94,62 + 4,44 detik, dan stabilitas yang baik dengan pengujian freeze-thawing 6 siklus. Stigmasterol yang terdisolusi selama 120 menit adalah 79,54 + 3,86 % dan mengikuti kinetika Weibull. Hasil penelitian menunjukkan bahwa formulasi S-SNEDDS mampu meningkatkan bioavailabilitas stigmasterol.

Purwoceng Gunung as aphrodisiac contains chemical components of steroid, essential oil, furanocoumarin, and vitamines, which are found in the radix. The steroid group consisted of sitosterol, stigmasterol (stigmasta-7, 16 dien-3-ol), and (stigmasta-7, 25 dien-3-ol). These chemical components make Purwoceng Gunung a traditional medicine to increase male vitality and fertility. Stigmasterol has a very low solubility in water, because it has semi-polar to non polar properties. The aim of this research were to know the optimal formula of S-SNEDDS ethanol extract Purwoceng Gunung with hazelnut oil as oil phase and the increase of stigmasterol dissolution percentage in vitro. Purwoceng Gunung radix extract was prepared by maceration method using 96% ethanol, then with n-hexane:chloroform (1:1). The extract was analyzed by stigmasterol content with HPLC. The SNEDDS formula used is hazelnut oil 15.00 %w/w, Tween 80 55.76% w/w, Croduret® 9.84% w/w, and propylene glycol 19.40% w/w. Maltodextrin is used as solidifying agent using spray drying method. S-SNEDDS is characterized by droplet size and particle distribution, angle of repose, bulk density, tapped density, drug content, powder morphology, stability, emulsification time, and dissolution test in vitro and its release kinetics. Purwoceng Gunung radix extract with yield of 9.61% contains stigmasterol with content of 11.96 + 0.93% w/w, thick texture, dark greenish brown, bitter taste and distinctive Purwoceng odor, moisture content 3.53 + 0.54, and heavy metal content Pb (<0.03 mg/kg), Cd (<0.01 mg/kg), and Hg (22.93 μg/kg). Water content test results and heavy metal content meet the requirements stipulated by BPOM RI. Extract loading in SNEDDS 150 mg/g with a particle size of 21.50 nm. S-SNEDDS contains stigmasterol with content 4.14 + 0.86 w/w, has poor flow properties, emulsification time 94.62 + 4.44 second, and good stability with freeze-thawing test 6 cycle. The stigmasterol which was dissolved in 120 minutes was 79.54 + 3.86% and followed the Weibull kinetics. The results showed that the S-SNEDDS formulation was able to increase the bioavailability of stigmasterol.
