STUDI HISTOLOGIS DAN INDEKS GONADOSOMATIK PERKEMBANGAN GONAD IKAN SIDAT (Anguilla bicolor bicolor McClelland, 1844)

Budidaya ikan sidat (Anguilla bicolor bicolor) di Indonesia masih terbatas hingga tahap pembesaran, karena belum dapat dipijahkan dikolam budidaya. Penelitian ini bertujuan untuk mengetahui korelasi antara nilai Index Gonadosomatik (IGS) terhadap panjang tubuh, berat total ikan dan berat gonad serta membandingkan struktur histologi gonad betina, undifferentiated dan jantan. Penelitian ini menggunakan 14 ikan sidat (Anguilla bicolor bicolor). Semua ikan diukur panjang dan berat total tubuhnya, gonad diisolasi dan ditimbang serta dibagi menjadi gonad kranial, medial dan kaudal. Preparat histologi gonad dibuat dengan pewarnaan Hematoxylin-eosin (HE) untuk deskripsi komparatif, menghitung jumlah oosit tahap oogonium, oosit primer, kortikal alveoli, vitellogenik dan oosit matang di gonad kranial, medial dan kaudal serta pengukuran keliling folikel kortikal alveoli, vitellogenik dan oosit matang pada ikan sidat betina. Pewarnaan Mallory Acid Fuchsin (MAF) digunakan untuk melihat susunan jaringan ikat gonad. Analisis data dilakukan secara kualitatif dengan deskripsi komparatif antara struktur histologi gonad betina, undifferentiated dan jantan. Analisis kuantitatif dilakukan dengan uji Two-way ANOVA, uji korelasi sederhana (Bivariate correlation) dan uji Regresi linier ganda dalam software SPSS ver.16. Hasil pengamatan struktur histologi gonad menunjukkan bahwa 14 ikan sidat (Anguilla bicolor bicolor) terdiri dari 7 ikan betina, 3 undifferentiated dan 3 jantan. Hasil analisis menunjukan nilai IGS ditentukan oleh panjang tubuh, berat tubuh dan berat gonad ikan sidat. Berat tubuh adalah faktor yang paling berpengaruh terhadap nilai IGS. Preparat histologis gonad betina disusun oleh oogonium, oosit primer, kortikal alveoli, vitellogenik dan oosit matang. Gonad undifferentiated disusun oleh sel PGC (Primordial Germ Cell) dan gonad jantan disusun oleh sel-sel spermatogenik, sel leydig dan sel sertoli.

Cultivation of tropical eel (Anguilla bicolor bicolor) in Indonesia hasn't grown well because it can't be spawned outside its habitat. This research aim to study the correlation of Gonadodomatic Index (GSI) with body lenght, body weight, gonad weight of tropical eel. The second aim is to compare the histological structure between female, undifferentiated and male gonad. This research use 14 tropical eels. Eel's Body lenght and body weight are measure then gonad is preserved and divide become cranial, medial and caudal part. Gonad histology are stained with Hematoxylin-eosin (HE) to make comparison, calculating the oogonium, primary oocyte, cortical alveoli, vitellogenic and mature oocyte also calculating the perimeter of cortical alveoli, vitellogenic and mature oocyte in female gonad. Mallory Acid Fuchsin (MAF) staining is used for identification the gonad connective tissue. The data were analyzed for both qualitatively and quantitatively. Qualitative analysis was conducted by descriptive comparative between histology of female, undifferentiated, and male gonad, while quantitative analysis performed by Two-way ANOVA, Bivariate correlation and Multiple linear regression analysis in SPPS ver.16. Based on observation from 14 tropical eels (Anguilla bicolor bicolor) gonad histology show that 4 eels are female, 3 undifferentiated and 3 male. GSI value is determined by body lenght, body weight and gonad weight. Body weight is the most influent factor. Female gonad consist of oogonium, primary oocyte, cortical alveoli, vitellogenic and mature oocyte. Undifferentiated gonad consist of PGC (Primordial Germ Cell). Male gonad consist of spermatogenic cells, leydig cell and sertoli cells.
