Optimasi Material Co-processed Amilum Sagu – PVP K30 Sebagai Filler-Binder Pada Sediaan Tablet Asetosal

INTISARI Amilum sagu berpotensi sebagai bahan tambahan pada sediaan tablet, tetapi jarang digunakan karena memiliki sifat alir yang buruk. PVP K30 dikenal dalam industri farmasi sebagai bahan pengikat yang baik pada sediaan tablet dan dapat dikombinasikan dengan amilum sagu menjadi suatu material co-processed. Penelitian ini bertujuan untuk mengetahui proporsi optimum dari amilum sagu serta PVP K30 dan pengaruh variasi keduanya terhadap sifat fisik material co-processed dan sifat fisik tablet asetosal. Pembuatan delapan run material co-processed dilakukan menggunakan metode spray drying berdasarkan metode Simplex Lattice Design (SLD). Evaluasi sifat fisik material co-processed dilakukan untuk mendapatkan respon guna mencari proporsi optimum. Evaluasi sifat fisik material co-processed yang dilakukan meliputi penentuan diameter rata-rata partikel, uji kecepatan alir, kompaktibilitas, kompresiblitas, uji kecepatan penyerapan air, dan uji kerapuhan material. Penentuan proporsi optimum menggunakan Software Design Expert® versi 7.1.5. Material co-processed hasil optimasi kemudian digunakan sebagai filler-binder pada pembuatan tablet asetosal. Evaluasi sifat fisik tablet asetosal meliputi uji keragaman bobot, uji kekerasan, penentuan waktu hancur, dan uji kerapuhan tablet. Variasi proporsi amilum sagu dan PVP K30 berpengaruh terhadap sifat fisik material co-processed. Peningkatan komposisi amilum sagu dapat meningkatkan daya serap air, kompresibilitas dan kerapuhan material co-processed, sedangkan peningkatan komposisi PVP K30 dapat meningkatkan diameter rata-rata partikel, kompaktibilitas, dan kecepatan alir. Proporsi optimum material co-processed merupakan material co-processed dengan kombinasi amilum sagu sebesar 82,25% dan PVP K30 sebesar 17,75%. Pembuatan tablet asetosal dengan menggunakan filler-binder material co-processed hasil optimasi menghasilkan tablet asetosal dengan sifat fisik tablet meliputi keragaman bobot, kekerasan, waktu hancur dan kerapuhan memenuhi persyaratan yang ditetapkan.

ABSTRACT Sago starch is a potential excipient to tablet preparations, but it’s rarely used because it has poor flow properties. PVP K30 is known in the pharmaceutical industry as a good binding agent in tablet preparations and can be combined with sago starch into a co-processed material. This study aims to determine the optimum proportion of sago starch-PVP K30 and the effect of both variation on the physical properties of co-processed materials and the physical properties of acetosal tablets. The making of eight run co-processed materials was performed uses spray drying method based on Simplex Lattice Design (SLD) method. Evaluation of the physical properties of co-processed materials was performed to obtain responses to find the optimum proportions. Evaluation of the physical properties of co-processed materials includes determination of particle diameter, flowability rate test, compactibility, compressibility, water absorption rate test, and material fragility test. Determination of optimum proportion using Software Design Expert® version 7.1.5. The optimized co-processed material is then used as a filler-binder in the preparation of the acetosal tablets. Evaluation of the physical properties of acetosal tablets includes weight diversity test, hardness test, determination of disintegration time, and tablet fragility test. The variation in the proportion of sago starch and PVP K30 has an effect on the physical properties of co-processed material. Increased sago starch composition can increase water absorption, compressibility and fragility of co-processed materials, while increasing the composition of PVP K30 can increase the average particle diameter, compactibility, and flowability rate. The optimum proportion of co-processed material is a co-processed material with a combination of 82.25% sago starch and PVP K30 of 17.75%. Preparation of acetosal tablets by uses optimized material co-processed filler-binder result tablets of acetosal with physical properties of tablets including weight diversity, hardness, disintegration time and friability comply with specified requirements.
