EFEK EKSTRAK ETANOL DAUN BINAHONG (Anredera cordofolia (Ten.) Steen.) PADA GINJAL TIKUS WISTAR DENGAN LUKA DIABETES

Diabetes merupakan gangguan metabolik yang menyebabkan berbagai komplikasi penyakit lainnya. Komplikasi yang sering terjadi yaitu gangguan ginjal dan luka diabetes. Pemanfaatan tanaman Binahong (Anredera cordofolia (Ten.) Steen.) untuk menyembuhkan berbagai luka telah banyak dilakukan. Oleh sebab itu penelitian ini bertujuan untuk mngetahui efek pemberian ekstrak etanol daun binahong terhadap ginjal tikus dengan luka diabetes. Tikus wistar jantan berumur 3 bulan dibagi menjadi 3 kelompok (A,B dan C) masing masing 4 tikus. Tikus kelompok A (kontrol non diabetes) diinjeksi 0,1M sodium citrat buffer, sedangkan tikus kelompok B (kontrol diabetes) dan kelompok C diinjeksi streptozotocin 40mg/kgBB secara intraperitoneal. Satu minggu kemudian dilakukan pengecekan kadar gula darah dan dilakukan pembuatan luka eksisi di daerah punggung pada seluruh tikus. Tikus kelompok A dan B diberi 1 ml NaCMC 1% dua kali perhari secara peroral, sedangkan tikus kelompok C diberi 1 ml ekstrak etanol daun binahong dosis 300mg/kgBB dua kali perhari secara peroral. Perlakuan dilaksanakan selama 14 hari. Setelah 14 hari tikus dinekropsi untuk pengambilan ginjal. Ginjal ditimbang dan dimasukkan ke dalam formalin 10% untuk analisis histopatologi dengan pengecatan Hematoxylin Eosin (HE). Gambaran histopatologi ginjal tikus yang diberi ekstrak etanol daun binahong menunjukkan berbagai perubahan diantaranya terjadi hipertrofi glomerulus dan atrofi glomerulus, sedangkan ginjal tikus kontrol diabetes terjadi perubahan yang sama disertai endapan protein pada urinary space dan nefrosis tubulus. Data rasio berat ginjal terhadap berat badan menunjukkan tikus kontrol non diabetes (0,72±0,15), tikus kontrol diabetes (0,88±0,04) dan tikus diabetes yang diberi ekstrak etanol daun binahong (0,90±0,39). Hasil analisis menunjukkan tidak terdapat perbedaan yang signifikan (P>0,05). Kesimpulan, pemberian ekstrak etanol daun binahong tidak berpengaruh terhadap kerusakkan ginjal akibat diabetes dan rasio berat ginjal terhadap berat badan tikus keseluruhan.

Diabetes is a metabolic disorder that causes various complications of the other disease. The most common complications are renal failure and diabetic wound. The use of Binahong leaf (Anredera cordofolia (Ten.) Steen.) to heal many wound has been researched a lot. Therefore, this research was conducted to study the effect of binahong (Anredera cordofolia (Ten.) Steen.) ethanol leaves extracts on wistar rat kidney with diabetic wound. Wistar rat at 3 months of age were divided into 3 group and each group contain 4 rat. A group (non-diabetic controls rats) was injected 0.1 M sodium citrate buffer, meanwhile B group (diabetic control rats) and C group were injected streptozotocin (40mg/kg) intra peritoneally. One week later, a blood glucose check was performed and an excision wound are made in the back area of all rats. The rats in A and B groups were given 1 ml of 1% NaCMC orally twice a day, while rats in C group were given 1 ml of binahong leaf ethanol extract (300mg/kg) orally twice a day. The treatment was conducted for 14 days. After 14 days, the rat were necropsized. The kidneys weight are measured and inserted into formalin 10% for histopathological analysis using hematoxylin eosin (HE) staining. The analyze of renal histopathologic of the rats treated with ethanol extract of binahong ethanol leaves extracts showed various change including glomerular hypertrophy and glomerular atrophy, while the diabetic control rats showed similar change with protein accumulation in urinary space and tubular nephrosis. The data of ratio of its renal weight to body weight showed that non diabetic control rats (0,72±0,15), diabetic control rats (0,88±0.04) and diabetic rats given ethanol extract of binahong leaf (0,90 ± 0,39). The analysis result showed that there was no significant difference (P> 0,05). In conclusion, giving of binahong leaves extract did not affect kidney damage due to diabetes and ratio of renal weight to body weight of all rats.
