EKSTRAK URANG-ARING (Eclipta alba) SEBAGAI IMMUNOMODULATOR PADA TIKUS PUTIH (Rattus norvegicus) YANG DIBERIKAN SEBELUM INFEKSI TAKIZOIT Toxoplasma gondii

Toxoplasma gondii adalah parasit obligat intraseluler yang dapat menginfeksi manusia dan hewan di seluruh dunia. Penelitian ini bertujuan untuk mengetahui adanya respon imun humoral terhadap infeksi T. gondii pada tikus melalui uji serologis Latex Agglutination Test (LAT) menggunakan Pastorex-Toxo® kit serta mengetahui aktivitas fagositosis makrofag pada tikus yang diberi ekstrak urang-aring (E.alba) sebelum diinfeksi T.gondii yang mempunyai kandungan Wedelolactone sebagai immunomodulator. Dua belas ekor tikus Wistar dibagi menjadi empat kelompok yaitu kelompok kontrol positif, kontrol negatif, perlakuan A-1 dan A-2 . Semua kelompok diambil cairan acites serta serum untuk uji serologis LAT dengan Pastorex-Toxo® kit. Hasil uji serologis LAT pada tikus yang diberi perlakuan ekstrak Urang-aring sebelum diinfeksi takizoit T.gondii menunjukkan bahwa muncul respon imunitas humoral di hari ke-3 pada kelompok perlakuan A-1 dan A-2 sedangkan pada kelompok kontrol positif respon humoral baru muncul pada hari ke-6 dan kelompok kontrol negatif tidak muncul respon humoral. Hasil uji fagositosis menunjukan bahwa tikus yang diberi terapi dengan ekstrak urang-aring terdapat peningkatan presentase fagositosis makrofag sebesar 85,71%, kontrol negatif 0%, kontrol positif 50% dan indeks fagositosis sebesar 180,00%, kontrol negatif 0%, kontrol positif 122,22% serta bermakna signifikan secara statistik (p<0,005). Ekstrak Urang-aring (E.alba) dapat sebagai immunomoduator pada tikus putih (Rattus norvegicus) yang diberikan sebelum infeksi T.gondii

Toxoplasma gondii is obligate intracellular parasite which can infect human and animal around the world. This research aims to know the the existence of the humoral immune response to infection T. gondii on the rat through the serologis test Latex Agglutination Test (LAT) using Pastorex ® Toxo- kit as well as knowing the activity of macrophage phagocytosis in rat infected with T. gondii after given extract Ecliptia alba which has the content of Wedelolactone as immunomodulator. Twelve Wistar rats were divided into four groups, the positive control, negative control, treatment A-1 and treatment A-2. All group of rats are taken liquid acites and taken a serum that was used for the test serologis LAT uses Pastorex-kit ® Toxo. LAT serologic test result from the rats that given the extract Urang-aring before the infection of T.gondii tachyzoit shows that humoral immune response occur in the 3rd day from group A-1 and A-2 while the group of positive control shows that the humoral immune response occur in the 6th day and the group of negative control did not show any humoral immune response. Test results showed the phagocytosis that mice who were given therapy with extract of Ecliptia alba there is an increase in percentage of macrophage phagocytosis activity in the amount of 85,71%, negative control 0%, positive control 50% and phagocytosis index in the amount of 180,00%, negative control 0%, positive control 122,22% in statistically significant means (p<0,005). Urang-aring (E.alba) extract can be use as immunomodulatory in rats (Rattus norvegicus) that given before the infection of T.gondii.
