IMUNOPATOLOGIS IMUNOHISTOKIMIA STREPTAVIDIN BIOTIN UNTUK DETEKSI AVIAN INFLUENZA VIRUS PADA UNGGAS PETELUR DENGAN GEJALA KLINIS TORTICOLLIS DAN CURLED TOE PARALYSIS

Infeksi virus avian influenza (AIV) pada unggas petelur dapat menyebabkan gejala klinis torticollis dan curled toe paralysis serta lesi patologis anatomis berupa petechial hemorrhages dan foci necrotic hemorrhages saluran pencernaan. Pada infeksi AIV kasus lapangan, gejala klinis dan lesi patologis tersebut seringkali serupa dengan gejala klinis dan lesi patologis yang dijumpai pada unggas yang terinfeksi virus lain, seperti Newcastle disease virus (NDV), sehingga cukup menyulitkan dalam peneguhan diagnosis kedua virus tersebut. Uji imunopatologis imunohistokimia streptavidin biotin (IHK SB) merupakan suatu pendekatan uji bioteknologi berbasis antibodi untuk mendeteksi patogen dengan prinsip deteksi antigen pada spesimen jaringan. Penelitian ini bertujuan untuk mengaplikasikan IHK SB untuk mendeteksi AIV pada unggas petelur kasus lapangan dengan gejala klinis torticollis dan curled toe paralysis serta lesi patologis anatomis berupa petechial hemorrhages dan foci necrotic hemorrhages pada paru-paru dan saluran pencernaan. Sampel berasal dari 20 ekor ayam petelur komersial kasus lapangan dengan gejala klinis torticollis dan curled toe paralysis yang diduga terinfeksi AIV. Otak, paru-paru, dan duodenum dengan lesi patologis anatomis berupa petechial hemorrhages dan foci necrotic hemorrhages diambil dan dibuat sediaan histopatologis blok parafin dan diuji dengan IHK SB. Hasil IHK SB diamati dengan microscope digital camera system dan dianalisis secara deskriptif kualitatif. Hasil penelitian ini membuktikan, bahwa dengan uji IHK SB, antigen AIV dapat dideteksi pada otak (8 dari 11), paru-paru (5 dari 9), dan duodenum (8 dari 8). Berdasarkan hasil penelitian ini, dapat disimpulkan, bahwa IHK SB dapat diaplikasikan untuk deteksi AIV pada unggas petelur dengan gejala klinis torticollis dan curled toe paralysis dan dapat dikembangkan sebagai konfirmasi diagnosis AIV pada kasus lapangan.

Avian influenza virus (AIV) infection in laying birds can cause clinical signs of torticollis and curled toe paralysis and pathologic anatomic lesions of petechial and foci necrotic hemorrhages in respiratory and gastrointestinal tracts. In the field cases, clinical signs and pathologic anatomic lesions of AIV infection are similar with clinical signs and lesions in other infected poultry viruses, such as Newcastle disease virus (NDV), making it difficult to confirm the infection of both viruses. Immunopathological immunohistochemistry streptavidin-biotin (IHC SB) is an antibody-based test to detect the presence of pathogens, especially AIV in poultry with the principle of antigen detection in tissue specimens. This research aims to apply IHC SB to detect AIV infection in laying birds in the field cases with clinical signs of torticollis and curled toe paralysis and pathologic anatomic lesions, such as petechial and foci necrotic hemorrhages of the lungs and digestive tract. The samples were taken from 20 layer chickens with clinical signs of torticollis and curled toe paralysis suspected of being infected AIV. After being necropsied, then the brains, lungs, and duodenum with pathologic lesions of petechial and foci necrotic hemorrhages embedded in paraffin and tested for IHC SB. The IHC SB results were observed with a digital microscope camera system and analyzed descriptively qualitative. The results of the present study indicated that by applying the IHC SB, AIV antigens can be detected in the brains (8 of 11), lungs (5 of 9), and duodenum (8 of 8). Based on these results, it was concluded that the IHC SB can be applied to the detection of AIV in laying birds with clinical signs of torticollis and curled toe paralysis and IHC SB can be developed for confirmative diagnosis of AIV in the field cases.
