EFEK EKSTRAK ETANOL DAUN BINAHONG (Anredera cordofolia (Ten.) Steen.) PADA HATI TIKUS WISTAR DIABETES DALAM KONDISI LUKA EKSISI

Luka diabetes merupakan masalah kesehatan yang sering terjadi. Pemanfaatan daun binahong (Anredera cordofolia (Ten.) Steen.) diketahui dapat menurunkan kadar gula darah dan mempercepat kesembuhan luka. Oleh sebab itu penelitian ini bertujuan untuk mengetahui efek ekstrak etanol daun binahong terhadap hati tikus wistar diabetes dalam kondisi luka eksisi. Dua belas tikus wistar jantan umur 3 bulan dibagi 3 kelompok (A,B dan C) masing masing 4 tikus. Tikus kelompok A (kontrol non diabetes) diinjeksi 0,1M sodium citrat buffer, sedangkan tikus kelompok B (kontrol diabetes) dan kelompok C diinjeksi streptozotocin 40mg/kgBB secara intraperitoneal. Satu minggu kemudian dilakukan pembuatan luka eksisi di daerah punggung pada seluruh tikus. Tikus kelompok A dan B diberi 1 ml NaCMC 1% dua kali perhari secara peroral, sedangkan tikus kelompok C diberi 1 ml ekstrak etanol daun binahong dosis 300mg/kgBB dua kali perhari secara peroral. Perlakuan dilaksanakan selama 14 hari. Setelah 14 hari tikus ditimbang berat badannya kemudian dinekropsi untuk pengambilan hati. Hati dimasukkan ke dalam formalin untuk analisis histopatologi dengan pengecatan Hematoxylin Eosin (HE). Hasil penelitian menunjukkan bahwa penghitungan selisih berat badan tikus kontrol non diabetes (4,35�±9,91 gram) secara signifikan lebih rendah dibandingan tikus kontrol diabetes (77,17�±20,78 gram) dan tikus diabetes yang diberi ekstrak etanol daun binahong (68,22�±20,96 gram)(P<0,05). Berat relatif hati pada tikus kontrol non diabetes (2,64�±0,23%), tikus kontrol diabetes (3,15�±0,11%) dan tikus diabetes dengan ekstrak binahong (2,68�±0,39%) tidak menunjukan perbedaan yang signifikan (P>0,05). Gambaran histopatologi hati tikus yang diberi ekstrak daun binahong menunjukkan adanya degenerasi melemak di daerah centrolobuler sedangkan tikus diabetes menunjukkan fokal nekrosis dan akumulasi glikogen. Kesimpulan, pemberian ekstrak etanol daun binahong dapat menghambat penurunan berat badan,dan menghambat peningkatan berat hati akibat diabetes. Gambaran histopalogi hati menunjukkan adanya perbaikan karena sudah tidak ditemukan fokal nekrosis.

Diabetic wound is a common health problem. The use of binahong leaf (Anredera cordofolia (Ten.) Steen.) is known to decrease blood sugar levels, and accelerate the healing of wounds. Therefore, this research was conducted to study the effect of binahong (Anredera cordofolia (Ten.) Steen.) ethanol leaves extracts on diabetic wistar rat liver with excision wound. Twelve males wistar rat at 3 months of age were divided into 3 group of 4 rat each. Group A (non-diabetic controls rats) was injected 0.1 M sodium citrate buffer, meanwhile group B (diabetes control rats) and Group C were injected streptozotocin (40mg/kg) intra peritoneally. One week later excision wound was made in the back of the rats. Rats in the groups A and B were given 1 ml of 1% NaCMC orally twice a day, while rats in the group C were given 1 ml of binahong leaf ethanol extract (300mg/kg) orally twice a day. The treatment was conducted for 14 days. After 14 days, the rat were weighed and necropsized. Liver were taken and fixed with formalin 10% for histopathological analysis using hematoxylin eosin (HE) staining. The result showed that the body weight difference of non diabetic control rat (4,35�±9,91gram) were significantly lower than diabetic control rats (77,17�±20,78gram) and diabetic rats with binahong leaves ethanol extract (68,22�±20,96gram) (P<0,05). Relative liver weight of non diabetic control rats (2,64�±0,23%), diabetic control rats (3,15�±0,11%) and diabetic rats with binahong leaves ethanol extract (2,68�±0,39%) were not significantly different(P>0,05). Histopathologic figure of diabetic rat liver with binahong leaves ethanol extract was showed fatty degeneration in the centrolobuler area while the diabetic control rats was showed focal necrosis and accumulation of glycogen. Based on the result, the binahong leaves ethanol extract can inhibit weight loss,and inhibits the increase of heart weight due to diabetic. Liver histophatologic figure was showed improvement since there was no more focal necrosis.
