PENGARUH KOMBINASI FRAKSI TAK LARUT ETIL ASETAT EKSTRAK ETANOLIK HERBA SAMBILOTO (Andrographidis Paniculatae Herbae) DAN PEGAGAN (Centellae Asiaticae Herbae) TERHADAP AKTIVITAS SGPT DAN HISTOPATOLOGI HATI PADA TIKUS DIABETES MELITUS TIPE 2 TERINDUKSI STREPTOZOTOSIN

Sambiloto (Andrographis paniculata (Burm. f) Ness) dan pegagan (Centella asiatica (L). Urban) merupakan tanaman obat unggulan yang sedang dikembangkan sebagai obat tradisional salah satunya sebagai antidiabetes. Kondisi diabetes dapat mennyebabkan perubahan pada aktivitas SGPT dan histopatologi hati. Penelitian bertujuan untuk melihat pengaruh kombinasi fraksi tak larut etil asetat ekstrak etanolik herba sambiloto dan herba pegagan terhadap aktivitas SGPT serta histopatologi hati pada tikus diabetes melitus tipe 2 terinduksi streptozotosin. Jenis penelitian ini adalah eksperimental. Lima puluh enam tikus dibagi dalam 8 kelompok meliputi kelompok normal (K1); kontrol negatif (K2): diberi Na-CMC 0,5 % p.o; kontrol positif (K3): diberi glibenklamid 4,5 mg/kg BB p.o; fraksi tak larut etil asetat ekstrak etanolik herba sambiloto tunggal 1303 mg/kg BB p.o (K4); fraksi tak larut etil asetat ekstrak etanolik herba pegagan tunggal 1000 mg/kg BB p.o (K5); kombinasi fraksi tak larut etil asetat ekstrak etanolik herba sambiloto dan pegagan 912,1 mg/ kg BB : 300 mg/kg BB p.o (K6); kombinasi fraksi tak larut etil asetat ekstrak etanolik herba sambiloto dan pegagan 651,5 mg/kg BB : 500 mg/kg BB p.o (K7); kombinasi fraksi tak larut etil asetat ekstrak etanolik herba sambiloto dan pegagan 390,9 mg/kg BB : 700 mg/kg BB p.o (K8). Parameter yang diukur yaitu aktivitas SGPT dan histopatologi hati. Data dianalisis menggunakan uji statistik dengan taraf kepercayaan 95%. Hasil uji statistik menunjukkan penurunan yang signifikan tehadap aktivitas SGPT (p<0,05) pada K3, K4 serta dua kelompok kombinasi yaitu K7 dan K8 setelah diberi perlakuan selama 14 hari. Sementara pemberian fraksi tak larut etil asetat ekstrak etanolik herba sambiloto dan pegagan tidak memberikan pengaruh yang signifikan terhadap gambaran histopatologi hati.

Diabetes conditions can cause changes in ALT activity and liver histopathology. The study aims to look at the combined effect of the insoluble fraction of ethyl acetate extract ethanolic of sambiloto and centella herb against ALT activity and liver histopathology in type 2 diabetes mellitus rats induced by streptozotocin. This type of research is experimental. Fifty-six rats were divided into 8 groups include normal group (K1); negative control (K2): given Na-CMC 0.5% P.O; positive control (K3): given glibenclamide 4.5 mg / kg P.O; single dose of insoluble fraction of ethyl acetate extract ethanolic of sambiloto herb 1303 mg / kg P.O (K4); single dose of insoluble fraction of ethyl acetate extract ethanolic of centella herb 1000 mg / kg P.O (K5); combination dose of insoluble fraction of ethyl acetate extract ethanolic of sambiloto and centella herb 912.1 mg / kg: 300 mg / kg P.O (K6); combination dose of insoluble fraction of ethyl acetate extract ethanolic of sambiloto and centella herb 651.5 mg / kg : 500 mg / kg P.O (K7); combination dose of insoluble fraction of ethyl acetate extract ethanolic of sambiloto and centella herb 390.9 mg / kg: 700 mg / kg P.O (K8). The measured parameters are ALT activity and liver histopathology. Data were analyzed using statistical test with a level of 95%. Statistical analysis showed significant decrease in ALT activity (p <0.05) at the K3, K4 and two combination group, K7 and K8 after being treated for 14 days. Meanwhile, insoluble fraction of ethyl acetate extract ethanolic of sambiloto and centella herb do not give a significant effect on liver histopathology picture.
