PENGARUH VITAMIN D TERHADAP CEDERA TUBULUS, PROLIFERASI SEL TUBULUS, DAN EKSPRESI E-CADHERIN PADA MENCIT DENGAN CEDERA ISKEMIA/REPERFUSI

Latar Belakang: Cedera Iskemia/Reperfusi ginjal menginduksi terjadinya cedera tubulus, proliferasi sel, dan hilangnya integritas sel. Beberapa penelitian yang telah dilakukan menunjukkan bahwa vitamin D bersifat renoprotektif, dimana dapat mengurangi dampak buruk iskemia/reperfusi. Tujuan: Mengkaji pengaruh pemberian vitamin D terhadap cedera tubulus, proliferasi sel, dan ekspresi Ecadherin pada gagal ginjal akut sebagai efek cedera iskemia/reperfusi pada mencit. Metode: Klamp Pediculus Renalis dilakukan untuk menginduksi cedera iskemia/reperfusi pada mencit jantan galur Swisss sebanyak 15 ekor. Mencit dibagi menjadi 3 kelompok, Sham Operation (SO), cedera iskemia/reperfusi (IR7), dan cedera iskemia/reperfusi dengan diberi vitamin D (IR7D). Pewarnaan PAS digunakan untuk menilai skor cedera Tubulus, PCNA digunakan untuk mengamati proliferasi sel, dan ekspresi E-cadherin diperiksa dengan PCR. Data dianalisis menggunakan software SPSS versi 16. Hasil: Cedera iskemia/reperfusi menginduksi cedera tubulus, proliferasi sel, dan ekspresi E-cadherin. Vitamin D mengurangi cedera tubulus secara bermakna (p<0,05) dan meningkatkan proliferasi sel tubulus serta ekspresi E-cadherin. Kesimpulan: Vitamin D mengurangi cedera tubulus, meningkatkan proliferasi sel dan ekspresi E-cadherin. Kata Kunci: Vitamin D, cedera tubulus, proliferasi sel tubulus, ekspresi E-cadherin

Background: Kidney ischemia/reperfusion injury induces tubular injury, cell proliferation, and loss of cell integrity. Some studies have known that vitamin D is renoprotective, which can reduce the adverse effect of ischemia/reperfusion injury. Purpose: To elucidate the effect of vitamin D against tubular injury, cell proliferation, and the expression of E-cadherin in acute renal failure as the effects of the ischemia/reperfusion injury in mice. Method: Pedicle rela clamping injury done to induce ischemia/reperfusion injury in fifteen male mice with Swiss strain. Mice were divided into 3 groups, Sham Operation (SO), ischemia/reperfusion injury (IR7), and ischemia/reperfusion injury by being given vitamin D (IR7D). Pas staining was used to assess tubule injury score, PCNA was used to observe cell proliferation, and expression E-cadherin assessed by PCR. The data were analyzed by using SPSS version 16. Result:Ischemia/reperfusion injury induced tubular injury, cell proliferation, dan expression of Ecadheri. Vitamin D reduce tubular injury significantly (p<0,05) and increasing the tubular cell proliferation and also expression of E-cadherin. Conclusion: Vitamin D attenuates tubular injury, increases cell proliferation and expression of Ecadherin. Keyword: Vitamin D, tubular injury, expression of Ecadherin
