PENGARUH EKSTRAK BUNGA ROSELLA (Hibiscus sabdariffa L.) TERHADAP PERTUMBUHAN Candida albicans PADA PLAT GIGI TIRUAN THERMOPLASTIC NYLON

Gigi tiruan thermoplastic nylon yang selalu berkontak dengan mukosa rongga mulut dapat mempermudah perlekatan dan pertumbuhan Candida albicans. Jumlah Candida yang berlebihan dapat menyebabkan infeksi Candidiasis. Bunga rosella merupakan salah satu tanaman herbal yang mempunyai efek anti jamur. Penelitian bertujuan untuk mengetahui pengaruh ekstrak bunga rosella terhadap pertumbuhan Candida albicans pada plat gigi tiruan thermoplastic nylon . Bunga rosella dilakukan ekstraksi menggunakan metode maserasi dengan pelarut etanol 70%.Thermoplastic nylon berbentuk cakram dengan ukuran diameter 10mm dan tebal 2mm dibuat sebanyak 27 buah. Seluruh subjek penelitian direndam dalam saliva buatan selama satu jam kemudian dimasukkan kedalam suspense Candida albicans 108 CFU/mL selama 24 jam dengan suhu 370C kemudian dibagi menjadi tiga kelompok. Setiap kelompok direndam dalam ekstrak rosella 30%,60% dan akuades (kontrol) kemudian dicuci menggunakan akuades dan dipindahkan pada conical tube kemudian digetarkan dengan vortex mixer. Penanaman Candida albicans dilakukan dengan mengambil 0,1 ml kemudian dimasukkan dalam petri dan diinkubasi selama 48 jam dengan suhu 370C, selanjutnya dilakukan perhitungan koloni jamur dan dihitung angka jamur. Hasil data penelitian dilakukan uji ANAVA satu jalur dan LSD dengan taraf signifikansi 95%. Hasil uji ANAVA satu jalur menunjukkan terdapat perbedaan bermakna pada kelompok kontrol, ekstrak bunga rosella 30% dan 60% terhadap pertumbuhan Candida albicans(p<0,05). Hasil uji LSD menunjukkan adanya perbedaan yang bermakna antar semua kelompok perlakuan (p<0,05).Kesimpulan penelitian ini adalah terdapat pengaruh ekstrak bunga rosella terhadap pertumbuhan Candida albicans pada plat gigi tiruan thermoplastic nylon.

ABSTRACT Thermoplastic nylon denture which is always in contact with the oral mucosa may facilitate attachment and growth of Candida albicans. The overgrowth of Candida albicans can cause Candidiasis. Rosella flower is one of herb which has anti fungal effect. This study aims to determine the effect of Rosella flower extract against the growth of Candida albicans in thermoplastic nylon denture plates. Rosella flowers were extracted using 70% ethanol by maceration extraction method. The disc-shaped thermoplastic nylon of 10 mm diameter and 2 mm thickness were made as many as 27 pieces. The whole samples were soaked in saliva for one hour then put into a suspension of 108 CFU/mL Candida albicans for as long as 24 hours with the temperature of 37oC. The samples were then divided into 3 groups. Each group was soaked in rosella extracts of 30%, 60% and aquades (as a control). After being soaked, these samples were rinsed using aquades, transferred into conical tube then vibrated with a vortex mixer. The cultivation of Candida albicans were carried out by taking 10 ������µL of each sample and incubated for 48 hours at a temperature of 37oC. After that, Candida albicans were counted. The data were analyzed by One Way ANOVA and LSD test with significance level of 95%. The results show that the average numbers of fungi in control groups, 30% and 60% extracts were 101.77+1.92; 30.33+2.34; and 1.00+1.01 respectively. The results of One Way ANOVA test showed statistically significant differences in the growth of Candida albicans between control groups, 30% and 60% rosella extracts (p<0,05). The test LSD resulted a significant difference among all treatment groups (p<0.05). The conclusion of this study is that there is an effect of rosella flower extracts againts the growth of Candida albicans in thermoplastic nylon denture plates.
