PENGARUH KOMPOSISI MEMBRAN GELATIN KALSIUM KARBONAT TERHADAP PERSENTASE MUATAN VASCULAR ENDOTHELIAL GROWTH FACTOR (VEGF)

2016 | Skripsi | S1 PENDIDIKAN DOKTER GIGIINTISARI Kerusakan tulang dapat diperbaiki dengan memicu proses regenerasi melalui aplikasi teknologi rekayasa jaringan menggunakan kombinasi perancah dan growth factor. Perancah dapat dibuat menggunakan kombinasi gelatin dan kalsium karbonat, sedangkan growth factor dapat diperoleh dari Platelet Rich Plasma (PRP). Salah satu growth factor yang penting dalam regenerasi tulang adalah Vascular Endothelial Growth Factor (VEGF). Penelitian ini bertujuan untuk mengetahui pengaruh komposisi membran gelatin-kalsium karbonat terhadap persentase VEGF yang dapat dimuat dalam perancah. Dalam penelitian ini, uji persentase muatan VEGF dilakukan terhadap membran gelatin-kalsium karbonat dengan komposisi 6:4 dan 7:3, masing-masing 10 spesimen. Human PRP dipreparasi menggunakan teknik double centrifugation dan diaktivasi menggunakan CaCl2 2% sebelum dikontakkan dengan membran gelatin-kalsium karbonat. Pengontakan membran gelatin-kalsium karbonat dengan human PRP dilakukan selama 1 jam. Konsentrasi VEGF dalam human PRP diukur menggunakan human VEGF ELISA kit sebelum dan sesudah membran gelatin-kalsium karbonat dikontakkan dengan human PRP. Rerata persentase muatan VEGF pada komposisi 6:4 lebih tinggi dibandingkan komposisi 7:3. Hasil analisis menggunakan Independent Sample T-Test menunjukkan perbedaan persentase muatan pada komposisi 6:4 dan 7:3 tidak signifikan secara statistik. Kesimpulan dari penelitian ini adalah komposisi gelatin-kalsium karbonat 6:4 atau 7:3 tidak berpengaruh terhadap persentase muatan VEGF dari human PRP.
ABSTRACT Bone defect can be repaired by promoting regeneration process with tissue engineering technology application, using combination of scaffold and growth factor. Scaffold can be made by combining gelatin and calcium carbonate. Growth factor can be obtained from platelet rich plasma (PRP). One of growth factor, which is important in bone regeneration, is vascular endothelial growth factor (VEGF). The aim of this study was to determine the effect of gelatin-calcium carbonate membrane composition to the loading percentage of VEGF. In this study, the loading percentage of VEGF test was conducted on two groups of gelatin-calcium carbonate membrane with 6:4 and 7:3 compositions, on each ten specimens. Human PRP was prepared using double centrifugation technique and activated using CaCl2 2%, before being contacted to gelatin-calcium carbonate membrane. The contact between gelatin-calcium carbonate membrane and human PRP was conducted for one hour. VEGF concentration in human PRP was measured using human VEGF ELISA kit, before and after the contact between gelatin-calcium carbonate membrane and human PRP. The average value of loading percentage of 6:4 composition was higher than 7:3 composition. Analysis result using Independent Sample T-Test showed that the loading percentage difference, in 6:4 and 7:3 compositions, was not significant statistically. The conclusion of this study was 6:4 or 7:3 composition of gelatin-calcium carbonate has no effect to the loading percentage of VEGF from human PRP.
