AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL LABU KUNING (Cucurbita maxima) TERHADAP Staphylococcus aureus ATCC 25923

Latar belakang : Labu kuning (Cucurbita maxima) merupakan bahan makanan yang sering digunakan sebagai makanan pendamping ASI. Buah ini mengandung berbagai macam zat gizi seperti vitamin A, kalsium, fosfor, zat besi, lemak, protein, dan zat lainnya yang dibutuhkan untuk tumbuh kembang bayi. Selain itu, labu kuning juga mengandung senyawa flavonoid, tanin, dan saponin yang efektif melawan bakteri penyebab keracunan makanan. Salah satu gejala keracunan makanan adalah diare. Di Indonesia, diare menjadi penyebab kematian tertinggi pada bayi dan balita. Salah satu penyebab diare diakibatkan oleh Staphylococcus aureus. Berdasarkan permasalahan tersebut dan potensi antibakteri dari labu kuning maka dibutuhkan penelitian mengenai uji aktivitas antibakteri labu kuning terhadap Staphylococcus aureus. Tujuan penelitian : Mengetahui aktivitas antibakteri ekstrak etanol labu kuning (Cucurbita maxima) terhadap Staphylococcus aureus. Metode penelitian : Jenis penelitian ini adalah eksperimental dengan rancangan Postest only control design. Konsentrasi ekstrak yang digunakan meliputi 15.000 µg/ml, 8.000 µg/ml, 4.000 µg/ml, 2.000 µg/ml, 1.000 µg/ml, 500 µg/ml, 250 µg/ml serta menggunakan kontrol positif yang berisi media BHI steril dan bakteri serta kontrol negatif yang berisi media steril BHI dan NaCl sebagai pembanding. Penelitian dalam penelitian ini meliputi penelitian kadar hambat minimal (KHM). Jika hasil KHM menunjukkan hasil positif, maka akan dilanjutkan penelitian kadar bunuh minimal (KBM) untuk mengetahui aktivitas antibakteri labu kuning (Cucurbita maxima) terhadap Staphylococcus aureus. Hasil : Berdasarkan penelitian kadar hambat minimum (KHM), ekstrak etanol labu kuning (Cucurbita maxima) tidak poten untuk menghambat pertumbuhan Staphylococcus aureus, sehingga penelitian kadar bunuh minimum (KBM) tidak perlu dilakukan. Kesimpulan : Ekstrak etanol labu kuning (Cucurbita maxima) tidak memiliki aktivitas antibakteri terhadap Staphylococcus aureus.

Background: Pumpkin (Cucurbita maxima) is a food that is often used as complementary foods for breastfeeding. These fruit contains a variety of nutrients such as vitamin A, calcium, phosphorus, iron, fat, protein, and other substances needed for growth and development of babies. In addition, pumpkin also contains flavonoids, tannins and saponins that are effective against the bacteria that causes food poisoning. One of the symptoms of food poisoning are diarrhea. In Indonesia, the diarrhea becomes highest cause of death in infants and toddlers. One cause of diarrhea is Staphylococcus aureus. Based on these problems and antibacterial potency of pumpkin, it is necessary to do a research on the antibacterial activity of pumpkin against Staphylococcus aureus. Objective: to evaluate the antibacterial activity of ethanolic extracts of pumpkin (Cucurbita maxima) against Staphylococcus aureus. Methods: This study is an experimental study type design with post-test only control design. The extract used include 15.000 µg/ml, 8.000 µg/ml, 4.000 µg/ml, 2.000 µg/ml, 1.000 µg/ml, 500 µg/ml, 250 µg/ml and using a positive control containing BHI medium is sterile and bacteria as well as the negative control containing sterile BHI media and NaCl as a comparison. The observations in this study include observation of minimum inhibitory concentration (MIC). If the results of the MIC showed positive results, then continued observation of minimum bactericidal concentration (MBC) to determine the antibacterial activity of pumpkin (Cucurbita maxima) against Staphylococcus aureus. Results: Based on the observation of the minimum inhibitory concentration (MIC), the ethanolic extract of pumpkin (Cucurbita maxima) is not potent to inhibit the growth of Staphylococcus aureus. So observations minimum bactericidal concentration (MBC) is not necessary. Conclusion: The ethanolic extract of pumpkin (Cucurbita maxima) with a concentration 15.000μg/ml does not have antibacterial activity against Staphylococcus aureus.
