ESTIMASI SINTESIS PROTEIN MIKROBIA RUMEN BERDASARKAN EKSKRESI DERIVAT PURIN DALAM URIN KAMBING BLIGON YANG DIBERI PAKAN FERMENTASI BERBASIS AMPAS TAHU DENGAN PENAMBAHAN BUFFER NaHCO3

Penelitian ini bertujuan untuk mengetahui pengaruh penambahan buffer NaHCO3 pada pakan fermentasi berbasis ampas tahu terhadap sintesis protein mikrobia rumen yang diestimasi berdasarkan ekskresi derivat purin (DP) dalam urin kambing Bligon. Ternak yang digunakan adalah 12 ekor kambing Bligon betina, berat badan rata-rata sebesar 23 sampai 26 kg yang ditempatkan pada kandang metabolisme dan dibagi menjadi 3 kelompok masing-masing berisi 4 ekor. Pakan yang diberikan adalah hijauan berupa rumput lapangan dan jerami kacang tanah serta pakan konsentrat fermentasi berbasis ampas tahu. Perbandingan pemberian pakan hijauan dan konsentrat yaitu 70:30. Perlakuan yang digunakan yaitu pakan fermentasi berbasis ampas tahu tanpa penambahan NaHCO3 sebagai kontrol (P0), penambahan NaHCO3 1,5% (P1), penambahan NaHCO3 3% (P2). Penelitian meliputi periode adaptasi selama 30 hari dan periode koleksi selama 7 hari. Sampel urin yang diperoleh dianalisis kandungan DP meliputi allantoin, asam urat, xanthin dan hipoxanthin. Data pH pakan fermentasi, kadar dan total ekskresi DP, bahan organik tercerna (BOT), estimasi sintesis protein mikrobia (EMNS), digestible organic matter in rumen (DOMR), dan efisiensi sintesis protein mikrobia dianalisis menggunakan analisis variansi dengan rancangan acak lengkap pola searah kemudian dilanjutkan dengan uji Duncan�s New Multiple Range Test. Hasil penelitian menunjukkan bahwa penambahan NaHCO3 memberikan pengaruh yang sangat nyata (P<0,01) terhadap pH pakan fermentasi, serta memberikan pengaruh yang nyata (P<0,05) terhadap kadar DP, total ekskresi DP dengan nilai pada P0, P1, P2 yaitu 2230,35 µmol/ekor/hari; 2474,51 µmol/ekor/hari; 2506,20 µmol/ekor/hari, dan terhadap nilai EMNS dengan nilai pada P0, P1, P2 yaitu 1,8 g N/hari; 2,01 g N/hari; 2,04 g N/hari. Kesimpulan dari penelitian ini adalah penambahan NaHCO3 sebesar 1,5% pada pakan konsentrat berbasis ampas tahu dapat meningkatkan pH pakan fermentasi, kadar dan total ekskresi derivat purin serta nilai EMNS pada kambing Bligon namun tidak berpengaruh terhadap efisiensi sintesis protein mikrobia.

The aim of this study was to investigate ruminal microbe protein synthesis through purine derivatives excretion on Bligon goat fed with fermented tofu waste supplemented with NaHCO3 as buffer. Twelve female Bligon goats whose 23 until kg 26 of body weight average were placed individually on metabolism stalls and divided into 3 groups (4 goats each). All goats received forages and fermented concenctrate mixed diet. The ratio of forages:concentrate is 70:30. Forages were consisted of field grass dan peanut straw, while rice brain and pollard were used as concentrate, while tofu waste was used as main concentrate. Dietary treatments were conducted by supplementing diet with NaHCO3 buffer : 0%; 1,5%, and 3% (based on as fed). Research was conducted for 37 days which includes feeding adaptation for 30 days. At the last 7 days, feed residue, feces, and also urine were collected everyday for futher chemical analysis. Moreover, urine was collected as total urine which was produced on 24 hours. Urine were further used for allantoin, uric acid, xanthin, and hypoxanthin analyses. All data were analyzed using one way ANOVA design followed by Duncan's Multiple Range Test. Suplementation of NaHCO3 significantly increased fermented tofu waste pH value (P value <0,05), concentration of purin derivatives, total excretion of purine derivatives, and EMNS (estimated microbial protein synthesis) (P value <0,05). The amount of purine derivatives (µmol/unit/day) on each treatment P0, P1, P2 were 2230,35; 2474,51; 2506,20 with EMNS (g N/d) were 1,8; 2,01; and 2,04. Total of purine derivatives excretion based on body weight metabolism, DOMR (digestible organic matter on rumen), and EMNS/DOMR (efficiency of microbial protein synthesis) were not affected by NaHCO3 supplementation. This study can be concluded that supplementation of NaHCO3 1,5% on fermented tofu waste increased concentration of purine derivatives, total excretion of purine derivatives, and EMNS on Bligon goat but non significant on efficiency of microbial protein synthesis.
