PENGARUH PENAMBAHAN LEVEL KONSENTRASI SUKROSA DALAM MEDIA PENGENCER SUSU SKIM TERHADAP KUALITAS SPERMA KAMBING PERANAKAN ETTAWA SETELAH PENYIMPANAN PADA SUHU 5 DERAJAT CELCIUS

2016 | Skripsi | S1 ILMU DAN INDUSTRI PETERNAKANPenelitian ini bertujuan untuk mengetahui kualitas sperma kambing Peranakan Ettawa (PE) yang diencerkan dengan susu skim dan penambahan sukrosa dengan level konsentrasi yang berbeda kemudian di simpan pada suhu 5 derajat Celcius selama 3 jam. Bahan pengencer yang digunakan kombinasi susu skim dengan level konsentrasi sukrosa 0 persen sebagai kontrol, 1 persen (S1), 2 persen (S2), 3 persen (S3), 4 persen (S4). Parameter yang diamati dalam penelitian ini adalah kualitas sperma kambing PE secara makroskopis meliputi volume, warna, konsistensi, derajat keasaman (pH), dan secara mikroskopis meliputi motilitas, viabilitas dan abnormalitas spermatozoa setelah penyimpanan pada suhu 5 derajat Celcius. Sperma yang telah diencerkan dan disimpan dalam lemari es dengan temperatur plus minus 5 derajat Celcius selama 3 jam sukrosa 0 persen memiliki motilitas 64,4 plus minus 2,96 persen, viabilitas 68,87 plus minus 3,38, dan abnormalitas 0,5 plus minus 0,00, sukrosa 1 persen memiliki hasil motilitas 70,6 plus minus 6,50, viabilitas 71,95 plus minus 4,09, dan abnormalitas 0,6 plus minus 0,22, sukrosa 2 persen memiliki hasil motilitas 63,8 plus minus 14,51, viabilitas 73,86 plus minus 3,32, dan abnormalitas 0,5 plus minus 0,00, sukrosa 3 persen memiliki hasil motilitas 67,2 plus minus 10,37, viabilitas 71,75 plus minus 4,59, dan abnormalitas 0,8 plus minus 0,27, sukrosa 4 persen memiliki hasil motilitas 66 plus minus 6,28, viabilitas 68,15 plus minus 1,96, dan abnormalitas 0,5 plus minus 0,00. Berdasarkan penelitian yang dilakukan dapat disimpulkan bahwa pengencer susu skim dan penambahan sukrosa dapat digunakan untuk pengenceran sperma.
This research aimed to determine the effect of addition level concentration of sucrose into diluent skim milk medium on ettawa cross bred goat sperm quality after storage at temperature 5 degree celcius. The used various skim milk combinations with addition of sucrose concentration level of 0 percent as control, 1 percent (S1), 2 percent (S2), 3 percent (S3), 4 percent (S4) were used as dilutent. The quality of the goat sperm included macroscopic (volume, color, consistency, acidity); and microscopic (motility, viability, and abnormalities) were eaxamined after stored at 5 degree celcius. Sperm that had been diluted and stored in a refrigerator at temperature 5 degree celcius for 3 hours, showed 64.4 plus minus 2.96 percent, 70.6 plus minus 6.50 percent, 63.8 plus minus 14.51 percent, 67.2 plus minus 10.37 percent, 66 plus minus 6.28 percent of motility for 0 percent, 1 percent, 2 percent, 3 percent, 4 percent concentration of sucrose respectively. The viability were 68.87 plus minus 3.38 percent, 71.95 plus minus 4.09 percent 73.86 plus minus 3.32 percent, 71.75 plus minus 4.59 percent, 68.15 plus minus 1.96 percent for 0 percent, 1 percent, 2 percent, 3 percent, 4 percent concentration of sucrose respectively. The abnormalitas showed the significant different between 1 percent (0.6 plus minus 0,22 percent) and 0 percent (0.5 plus minus 0,00 percent), 2 percent (0.5 plus minus 0,00 percent), 3 percent (0.8 plus minus 0,27 percent), 4 percent (0.5 plus minus 0,00 percent). Based on this research can be concluded that the dilution of skim milk and sucrose can be used for sperm dilution.
