EFEK OBAT BATUK X TERHADAP PENINGKATAN SEKRESI MUKUS PADA TRAKEA MENCIT

Fungsi batuk sebagai pertahanan respiratorik dengan cara membawa keluar mukus saluran respiratorik yang berlebihan. Sekresi mukus ditingkatkan dengan ekspektoran. Salah satu produk yang diklaim sebagai ekspektoran oleh produsennya adalah Obat Batuk X. Penelitian ini bertujuan mengetahui efek pemberian sirup Obat Batuk X terhadap peningkatan sekresi mukus pada mencit jantan galur Balb/c dengan bobot 20-30 g pada kondisi fisiologi normal tanpa induksi. Mencit dibagi dalam lima kelompok. Kelompok I sebagai kontrol negatif dipejani dengan 0,5% Na-CMC. Kelompok II sebagai kontrol positif dipejani dengan 156 mg/kgBB glyceryl guaiacolate. Kelompok III, IV, dan V masing-masing dipejani dengan 2,92 mL/kgBB, 5,85 mL/kgBB dan 11,7 mL/kgBB Obat Batuk X. Tiap kelompok dipejani empat hari berturut-turut. Satu jam setelah hari ke-4, semua kelompok diinjeksi dengan 5% phenol red intraperitoneal. Konsentrasi mukus pada trakea mencit direpresentasikan dengan konsentrasi tracheal phenol red setelah perlakuan. Hasil penelitian menunjukkan bahwa konsentrasi tracheal phenol red cenderung meningkat seiring dengan peningkatan dosis Obat Batuk X, walaupun secara statistik tidak ada perbedaan bermakna antara konsentrasi tracheal phenol red pada kelompok yang dipejani Obat Batuk X dosis 2,92 mL/kgBB, 5,85 mL/kgBB dan 11,7 mL/kgBB dibandingkan dengan kelompok kontrol (p=0,453). Hal ini bermakna bahwa mukus yang disekresikan semua kelompok relatif sama banyak dan tidak ada kelompok perlakuan yang memiliki efek ekspektoran pada mencit dengan kondisi fisiologi normal tanpa induksi. The function of cough is respiratoric barrier that release excessive mucus. The release of mucus is increased by expectorant. Obat Batuk X is claimed as expectorant by its producer. This study aimed to determine the effect of Obat Batuk X on enhancement of mucus secretion in mice's trachea (Balb/c, 20-30 g, in physiological condition without induction). Mice were devided into 5 groups: negative control (0,5% Na-CMC p.o.), positive control (156 mg/kgBW glyceryl guaiacolate p.o.), and 3 doses of Obat Batuk X (2,92 mL/kgBW, 5,85 mL/kgBW, 11,7 mL/kgBW p.o.). Mice were injected for 4 days. An hour after injection at fourth day, mice were injected with phenol red 500 mg/kgBW i.p and sacrificed 30 minutes later. The mucus's concentration is represented by concentration of tracheal phenol red output. The result of this study showed that concentration of tracheal phenol red output was increase as the enhancement of Obat Batuk X's dose, although there were not difference on tracheal phenol red output between Obat Batuk X groups and control groups (p=0,453). This result showed that all of mice secreted mucus in similar amount and there were not expectorant effect in this study (physiological condition without induction).
