AKTIVITAS IMUNOMODULATOR KOMBINASI EKSTRAK ETANOLIK UMBI KELADI TIKUS (Typhonium flagelliforme (Lodd.) Blume) DAN MENIRAN (Phyllanthus niruri L.) TERHADAP FAGOSITOSIS MAKROFAG, PROLIFERASI LIMFOSIT, DAN TITER ANTIBODI SECARA IN VIVO

Pemanfaatan bahan alam sebagai agen imunomodulator mampu memenuhi kebutuhan manusia dalam memperoleh sistem kekebalan tubuh yang optimal. Berdasarkan penelitian yang telah dilakukan sebelumnya, umbi keladi tikus (Typhonium flagelliforme (Lodd.) Blume) dan meniran (Phyllanthus niruri L.) memiliki aktivitas yang berpotensi sebagai agen imunomodulator. Walaupun demikian, daya imunomodulator kombinasi umbi keladi tikus dan meniran belum pernah diuji. Daya imunomodulator kombinasi ekstrak umbi keladi tikus dan ekstrak meniran diuji melalui proses pemejanan terhadap hewan uji selama 17 hari dengan diinduksi vaksin Hepatitis B. Hewan uji yang digunakan adalah tikus jantan galur Sprague-Dawley berusia 8 minggu dengan bobot ±100 gram sebanyak 36 ekor dan dibagi ke dalam 6 kelompok, yaitu DMSO,ekstrak keladi tikus tunggal, ekstrak meniran tunggal, kombinasi ekstrak keladi tikus 125 mg/kg BB-meniran 25 mg/kg BB, ekstrak keladi tikus 250 mg/kg BB-meniran 50 mg/kg BB, dan ekstrak keladi tikus 500 mg/kg BB-meniran 100 mg/kg BB. Pada hari ke-18 dilakukan pengambilan darah melalui plexus retroorbitalis dan pembedahan. Selanjutnya, dilakukan uji aktivitas fagositosis makrofag, proliferasi limfosit, dan titer antibodi. Data yang diperoleh menunjukkan bahwa kombinasi ekstrak keladi tikus dan meniran tidak mempengaruhi fagositosis makrofag yang merepresentasikan respon imun nonspesifik. Kombinasi ekstrak umbi keladi tikus 125 mg/kg BB-meniran 25 mg/kg BB, ekstrak umbi keladi tikus 250 mg/kg BB-meniran 50 mg/kg BB, dan ekstrak umbi keladi tikus 500 mg/kg BB-meniran 100 mg/kg BB mampu meningkatkan proliferasi limfosit, tapi tidak terdapat perbedaan yang signifikan antar variasi kelompok dosis uji tersebut dalam meningkatkan proliferasi limfosit. Kombinasi ekstrak umbi keladi tikus dan meniran tidak mempengaruhi produksi imunoglobulin G. Oleh karena itu, kombinasi ekstrak umbi keladi tikus-meniran terhadap respon imun spesifik hanya mempengaruhi respon imun selular, tetapi tidak mempengaruhi respon imun humoral.

Utilization of natural materials as immunomodulator agent is an answer of human needs to get the best immune system. Based on recent research, umbi keladi tikus (Typhonium flagelliforme (Lodd.) Blume) and meniran herb (Phyllanthus niruri L.) have an activity as immunomodulatory agent. Although, immunomodulatory activity of keladi tikus and meniran as a combination have never been researched. Immunomodulator activity of combination umbi keladi tikus and meniran is tested by animal testing that treated for 17 days and inducted by Hepatitis B vaccine. Thirty-six of SpragueDawley rats about 8 weeks and ±100 grams of weight is divided into six groups. They are DMSO group, keladi tikus extract group, meniran extract group, 125 mg/kg of body mass keladi tikus extract-25 mg/kg of body mass meniran extract group, 250 mg/kg of body mass keladi tikus extract-50 mg/kg of body mass meniran extract group, and 500 mg/kg of body mass keladi tikus extract-100 mg/kg of body mass meniran extract group. On the eighteenth day, blood sampling is took from the plexus retroorbitalis and sacrificed. Then, activity of macrophage phagocytosis, proliferation of lymphocyte, and antibody titre is tested. Data shows that combination of extract keladi tikus and meniran is not affecting phagocytosis of macrophage that represent non specific immune response. Combination of extract keladi tikus and meniran with dose 125 mg/kg of body mass keladi tikus extract-25 mg/kg of body mass meniran extract, 250 mg/kg of body mass keladi tikus extract-50 mg/kg of body mass meniran extract, and 500 mg/kg of body mass keladi tikus extract-100 mg/kg of body mass meniran extract were able to increase proliferation of lymphocyte, but there are no significant difference among groups. Combination of extract keladi tikus and meniran did not affecting production of immunoglobulin. Therefore, combination of extract keladi tikus and meniran only affecting cellular immune response, but did not affecting humoral immune response.
