Dinamika folikel ovulasi dan korpus luteum setelah sinkronisasi estrus pada sapi perah peranakan Friesian Holstein

2008 | Disertasi |Performan reproduksi sapi perah peranakan Friesian Holstein (PFH)akseptor inseminasi buatan umumnya masih rendah. Tujuan penelitian ini adalah: 1). Mengetahui dinamika perkembangan folikel dominan dan korpus luteum, serta profil progesteron plasma selama siklus estrus pada sapi PFH, 2). Mengetahui dinamika perkembangan folikel ovulasi, korpus luteum dan hormon progesteron setelah sinkronisasi dengan progesteron dan prostaglandin F2(simbol)(PGF2(simbol))(simbol)untuk inseminasi buatan terjadwal (timed artificial insemination) pada sapi PFH. Empat penelitian telah dilakukan, yaitu: akurasi diagnosa ultrasonografi transrektum untuk pemeriksaan struktur ovaria (penelitian 1), dinamika perkembangan folikel dominan selama siklus estrus (penelitian 2), perkembangan folikel ovulasi setelah erlakuan sinkronisasi estrus dengan implan progesteron intravagina (penelitian 3), dan perkembangan folikel ovulasi setelah perlakuan sinkronisasi estrus dengan PGF2(simbol) (penelitian 4). Hasil dari penelitian: 1). Pemeriksaan ultrasonografi ovaria transrektum terbukti mempunyai akurasi dan reliabilitas tinggi untuk identifikasi folikel dominan maupun korpus luteum. 2). Dinamika folikel selama siklus estrus pada sapi PFH hanya mempunyai 2 gelombang pekembangan folikel dominan. 3). Profil kadar progesteron plasma paralel dengan dinamika perkembangan korpus luteum selama siklus estrus. 4). Insersi implan intravagina controlled internal drug release (CIDR) menyebabkan kenaikan kadar progesteron plasma, pencabutan implan setelah 9 hari menimbulkan penurunan tiba-tiba kadar progesteron plasma, diikuti perkembangan sangat cepat folikel dominan, estrus dan ovulasi. Pemberian gonadotrophin releasing hormone (GnRH) pada saat pengambilan implan CIDR menambah laju perkembangan folikel ovulasi. 5). Pemberian PGF2(simbol) akan menyebabkan regresi korpus luteum diikuti dengan penurunan kadar progesteron plasma, perkembangan folikel dominan secara cepat, estrus dan ovulasi. 6). Protokol sinkronisasi estrus dengan implan CIDR yang paling sesuai untuk diaplikasikan pada sapi perah PFH adalah insersi intravagina 9 hari diikuti dengan penyuntikan GnRH 250 (simbol)g pada saat pengambilan implan. 7). Protokol sinkronisasi estrus dengan sediaan PGF2(simbol) terbaik untuk sapi PFH adalah pemberian PGF2 (simbol) diikuti dengan injeksi GnRH 250 (simbol)g 2 hari kemudian.
Reproductive performance of local Friesian cows as artificial insemination acceptors generally is still low. The present study aimed to: 1). Understand the dynamics of dominant follicle and corpus luteum development, as well as plasma progesterone profile throughout the estrous cycle in local Friesian dairy cows. 2). Know the dynamics of dominant follicle and corpus luteum development, as well as plasma progesterone profile after estrus synchronization treatment using progesterone intravaginal implant and prostaglandin F2(symbol) (PGF2(symbol)) for timed artificial insemination in local Friesian dairy cows. Four experiments had been carried out: the accuracy of transrectal ultrasonography for examining of cattle ovarian structures (experiment 1), the dynamics of dominant follicular development throughout the estrous cycle (experiment 2), the ovulatory follicular development after estrus synchronization treatment using progesterone intravaginal implant controlled internal drug release (CIDR) (experiment 3), and the ovulatory follicular development after estrus synchronization treatment using PGF2(symbol) (experiment 4). The results indicated that: 1). Transrectal diagnostic ultrasonography proved to have high precision and reliability for identification of the dominant follicle and corpus luteum. 2). Follicular dynamics throughout the estrous cycle in local Friesian dairy cattle had only two waves of development. 3). The plasma progesterone profiles were concomitant with the dynamics of corpus luteum during estrous cycle. 4). Intravaginal insertion of CIDR cause increased in plasma progesterone concentration and its withdrawal after 9 days resulting in sudden decreased followed by rapid development of the ovulatory follicle dominant and then ovulation. Intravaginal implant CIDR also diminished development of the existing corpus luteum. 5). Administration of PGF2(symbol) will cause regression of corpus luteum followed by decreased in plasma progesterone, rapid development of dominant follicle and eventually ovulation. 6). The most suitable estrus synchronization protocol using CIDR intravaginal implant for local Friesian cows is 9 day insertion followed by intramuscular injection of GnRH 250 (symbol)g at the time of implant removal. 7). The PGF2(symbol)(symbol)best protocol for local Friesian dairy cows is administration of the agent followed by intramuscular injection of GnRH 250 (symbol)g at 2 days later.
