Pengaruh ekstrak air daun Ceplikan (Ruellia tuberosa L) terhadap glukosa darah dan gambaran histologis pankreas tikus putih (Rattus norvegicus) diabetes melitus

Latar Belakang : Kontrol glukosa darah yang efektif dan aksi antioksidan merupakan kunci dalam mencegah diabetes dan komplikasinya, banyak tanaman herbal yang memiliki kedua efek tersebut. Daun Ceplikan (Ruellia tuberosa L.) merupakan salah satu obat tradisional yang secara empiris digunakan untuk menurunkan kadar glukosa darah. Selain senyawa antioksidan yang terkandung dalam Daun Ceplikan (Ruellia tuberosa L.) kemungkinan ada senyawa yang belum terdeteksi sehingga yang menjadi masalah disamping dapat menurunkan kadar glukosa darah, apakah juga mempunyai efek samping terhadap sel beta pankreas. Tujuan : Mengkaji pengaruh ekstrak air daun ceplikan (Ruellia tuberosa L.) terhadap kadar gula darah dan gambaran histologis pankreas pada tikus putih (Rattus norvegicus L) diabetes melitus. Metode Penelitian : Tiga puluh subjek tikus putih jantan galur wistar (Rattus norvegicus) umur 2-3 bulan dengan berat badan 150 – 200 gram dibuat diabetes dengan induksi aloksan dan secara randomisasi dibagi dalam 5 kelompok. Kelompok I adalah tikus diabetes diberi aquades, kelompok II diabetes diberi glibenklamid, kelompok III diabetes diberi ekstrak air daun ceplikan konsentrasi 1.6 mg, kelompok IV diabetes diberi ekstrak air daun ceplikan konsentrasi 3.2 mg, dan kelompok V diabetes diberi ekstrak air daun ceplikan konsentrasi 6,4 mg. Perlakuan diberikan secara oral setiap hari selama 30 hari. Kadar glukosa darah diukur pada hari 0, 3, 14 dan 30. Analisis statistik dengan uji repeated measures dan uji t-test. Hasil : Pemberian EADC dapat menurunkan kadar glukosa darah tikus DM. Penurunan maksimal pada ekstrak kadar 3,2 mg. Gambaran histologis pankreas tikus menunjukkan adanya perbaikan gambaran histologis pankreas ditandai dengan jumlah pulau langerhans masih banyak dan diameter pulau langerhans masih luas. Kesimpulan : Ekstrak air daun ceplikan dapat menurunkan kadar glukosa darah dan gambaran histologis pankreas tikus putih menunjukkan adanya perbaikan

Background: Effective control of blood glucose and antioxidant activities are key factors that prevent diabetes and its complications. There are lots of herbal plants that have both effects. Ruellia tuberosa L is are of traditional medicines which is empirically used to lower blood glucose level. A part from antioxidant compound contained in Ruellia tuberosa L it is possible there is other compound that has not been detected so that it raises a question whether there is side effect to pancreatic beta cells. Objective: To identify the effect of Ruellia tuberosa L extract to blood glucose level and pancreas histology description in white rats (Rattus norvegicus L.) with diabetes mellitus (DM). Method: Thirty subjects of Wistar strain male white rats (Rattus norvegicus) of 2-3 months old and of 150-200 grams weight were made diabetic with aloxan and randomly divided into 5 groups. Group I consisted of diabetic rats with aquadest, group II with glibenclamide, group III with extract of Ruellia tuberosa L 1.6mg, group IV with extract Ruellia tuberosa L 3.2mg, and group V with extract of Ruellia tuberosa L 6.4mg. Treatment was given orally per day within 30 days. Level of blood glucose was measured in day 0, 3, 4 and 30. Statistical analysis used repeated measures and t-test. Result: The supply of Ruellia tuberosa L extract could reduce level of blood glucose of DM rats. Maximal decreasing in extract 3,2 mg. Pancreas histological description of rats showed that there was improvement as indicated by greater quantity of Langerhans island and wider diameter of langerhans island. Conclusion: Ruellia tuberosa L extract could reduce level of blood glucose and Pancreas histological description of rats showed that there was improvement.
