Induksi Kalus Embriogenik pada Daun Eucalyptus pellita F. Muell

Eucalyptus pellita F. Muell merupakan salah satu tanaman dari Australia yang mudah tumbuh dan dibudidayakan pada iklim tropis maupun subtropis. Tanaman ini banyak dimanfaatkan oleh industri penghasil bahan baku kertas (pulp) karena batang tanaman ini mengandung berat jenis, persentase serat,kandungan selulosa, lignin dan kandungan ekstrak yang baik untuk produksi pulp. Produksi kertas di Indonesia masih sangat rendah dibandingkan dengan angka kebutuhan kertas. Perbanyakan tanaman ini secara konvensional seperti stek dan biji belum dapat menghasilkan bibit secara cepat dengan kualitas seragam seiring dengan tingginya permintaan bibit. Penelitian ini bertujuan untuk mengembangkan produksi bibit secara in vitro metode kultur jaringan melalui induksi embriogenesis somatik dari kalus (indirect somatic embryogenesis), menentukan medium dan kombinasi ZPT BA dan NAA terbaik untuk pembentukan kalus serta karakterisasi morfologi kalus yang terbentuk. Penelitian ini dilakukan dalam dua tahap yaitu perkecambahan biji untuk mendapatkan eksplan berupa daun secara in vitro dan induksi kalus dari eksplan. Induksi kalus dilakukan dengan mengkultur eksplan daun pada medium Murashige dan Skoog (MS) pada sembilan kombinasi ZPT BA dan NAA yaitu Ma (MS0); Mb (MS+ BA 0 mg/L + NAA 1 mg/L); Mc (MS+ BA 0 mg/L + NAA 2 mg/L); Md (MS+ BA 1,5 mg/L + NAA 0 mg/L; Me (MS+ BA 1,5 mg/L + NAA 1 mg/L); Mf (MS+ BA 1,5 mg/L + NAA 2 mg/L); Mg (MS+ BA 3 mg/L + NAA 0 mg/L); Mh (MS+ BA 3 mg/L + NAA 1 mg/L); Mi (MS+ BA 3 mg/L + NAA 2 mg/L). Hasil penelitian menunjukkan bahwa setelah ditumbuhkan selama 5 minggu terdapat empat perlakuan yang berhasil menginduksi terbentuknya kalus yaitu, Me, Mf, Mh, Mi dengan morfologi kalus kompak, berwarna kuning keputihan hingga kuning hijau keputihan yang tumbuh pada pangkal daun disekitar ibu tulang daun.Perlakuan terbaik dalam menginduksi kalus adalah kombinasi MS+ BA 1,5 mg/L + NAA 1 mg/L (Me).

Eucalyptus pellita F. Muell is a plant from Australia that easy to grow and cultivated in tropical and subtropical climates. This plant is widely used in paper industry because the stem of this plant contains specific gravity, fiber percentage, cellulose content, lignin and good extract content for pulp production. Paper production in Indonesia is still very limited compared to the number of paper needs.The high market demand for paper raw materials is not balanced with an increase in a seed sources. Conventional propagation of these plants such as cuttings and seeds has not been able to produce seedlings quickly with the same quality along with the high demand for seedlings. The objective of this study is to develop in vitro seed production method with the tissue culture technique, somatic embryogenesis through callus phase (indirect somatic embryogenesis), determine the best medium and combination of PGR�s (BA and NAA) on the formation of callus and the morphological character of the callus. This research was conducted in two step, seed germination and callus induction. The callus induction was carried out by culturing leaves explants on Murashige and Skoog (MS) medium with various combination of PGR�s which are Ma (MS0); Mb (MS+ BA 0 mg/L + NAA 1 mg/L); Mc (MS+ BA 0 mg/L + NAA 2 mg/L); Md (MS+ BA 1,5 mg/L + NAA 0 mg/L; Me (MS+ BA 1,5 mg/L + NAA 1 mg/L); Mf (MS+ BA 1,5 mg/L + NAA 2 mg/L); Mg (MS+ BA 3 mg/L + NAA 0 mg/L); Mh (MS+ BA 3 mg/L + NAA 1 mg/L); Mi (MS+ BA 3 mg/L + NAA 2 mg/L). The results showed that after being grown for 5 weeks there were four treatments that succeed in inducing callus formation namely, Me, Mf, Mh, Mi with compact morphology, whitish-yellow to whitish-green yellow growing at the base of the leaf around the leaf mother bones. The best treatment for inducing callus was a combination of MS + BA 1.5 mg / L + NAA 1 mg / L (Me).
