Efek Protektif Ekstrak Etanolik Bekatul Beras Hitam (Oryza sativa L. 'Sembada Hitam') Terhadap Sel NIH3T3 Yang Diinduksi Stres Oksidatif H2O2

Stressor endogen dan eksogen seperti metabolisme oksigen di mitokondria, radiasi, obat-obatan, dan polutan memicu stress oksidatif memiliki konsekuensi negatif sistem biologis yang mengarah pada berbagai patofisiologis dan mempengaruhi rentang hidup organisme. Berbagai metabolit sekunder yang terkandung di bahan alam seperti beras hitam diketahui mempunyai aktivitas antioksidan tinggi yang dapat mencegah terjadinya stress oksidatif. Penelitian ini bertujuan untuk mengkaji efek ekstrak etanolik bekatul beras hitam (Oryza sativa L. 'Sembada Hitam') terhadap sel NIH3T3 yang diinduksi H2O2. Fokus penelitian ini yaitu untuk mempelajari pengaruh ekstrak dalam menghambat sitotoksitas dan apoptosis sel NIH3T3 akibat induksi H2O2. Penelitian ini menggunakan kombinasi perlakuan H2O2 berbagai konsentrasi (50, 100, 150, 200, dan 300 uM) dan ekstrak bekatul beras hitam (EBBH) berbagai konsentrasi (7.8125; 15.625; 31.25; 62.5; 125; 250; 500, dan 1000 ug/mL). Data dianalisis menggunakan oneway ANOVA (p < 0.05) dilanjutkan dengan uji letak beda nyata Tukey HSD. Hasil menunjukkan bahwa EBBH pada konsentrasi 7.8125; 15.625; 31.25; 62.5; 125; 250; 500; dan 1000 ug/mL dapat mempertahankan viabilitas sel NIH3T3 di atas 80% dari paparan H2O2 konsentrasi 50 dan 100 uM selama 24 jam. Hal ini seiring dengan hasil uji apoptosis menggunakan double staining acridine orange-propidium iodide yang menunjukkan bahwa EBBH dapat menekan apoptosis khususnya pada kombinasi konsentrasi EBBH : H2O2 (62,5 ug/mL : 100 uM, 62,5 ug/mL : 200 uM dan 250 ug/mL:100 uM). EBBH pada konsentrasi 62,5 ug/mL terbukti paling efektif sebagai anti-apoptosis pada sel NIH3T3 induksi stress oksidatif H2O2.

Endogenous and exogenous stressors such as oxygen metabolism in mitochondria, radiation, drugs, and pollutants, which trigger oxidative stress, negatively affect biological systems. The effect leads to various pathophysiological conditions and affects the life span of organisms. Secondary metabolites in natural coumpound such as black rice have high antioxidant activity that can prevent oxidative stress. This study aimed to examine the effect of the ethanolic extract of black rice (Oryza sativa L. 'Sembada Hitam') bran on H2O2-induced NIH3T3 cells. The focus of this research is to evaluate the effect of the extract on cytotoxicity and apoptosis due to H2O2 induction. In this study, we used a combination of H2O2 exposure at various concentrations (50, 100, 150, 200 and 300 uM) and extract black rice bran (BRB) in various concentrations (7.8125; 15.625; 31.25; 62.5; 125; 250; 500, and 1000 ug/mL). The data were analyzed using one-way ANOVA (p < 0.05) followed by the Tukey HSD real difference test. Our results showed that extract of BRB at 7,8125 to 1000 ug/mL maintained NIH3T3 cell viability above 80% against 50 and 100 uM H2O2 exposure for 24 hours. These were in line with the results of the apoptosis test, which showed that the extract of BRB suppresses apoptosis. Especially the combination of BRB and H2O2 exposure at 62.5 ug/mL and 100 uM; 62.5 ug/mL and 200 uM, as well as 250 ug/mL and 100 uM, respectively. BRB pretreatment at the concentration of 62.5 ug/mL proved highly effective as an anti-apoptotic on H2O2-induced on NIH3T3 cells.
