EVALUASI PENGARUH KONFIGURASI REAKTOR DAN MEDIA IMOBILISASI TERHADAP POPULASI MIKROORGANISME DAN PRODUKSI BIOGAS DALAM PROSES PERURAIAN VINASSE SECARA ANAEROBIK TERMOFILIK

Penelitian ini bertujuan untuk meningkatkan produksi biogas melalui pemilihan konfigurasi reaktor dan media imobilisasi dalam rangka mengarahkan populasi mikroorganisme yang berperan dalam proses pembentukan metana dari vinasse pada kondisi termofilik. Penelitian ini terdiri atas empat tahapan eksperimen yaitu: 1) strategi start-up, 2) reaktor fixed-bed, 3) reaktor alir tangki berpengaduk (continuous stirred tank reactor/ CSTR) dengan zeolit, dan 4) reaktor fluidisasi anaerob. Kendala dalam proses termofilik adalah keterbatasan sumber inokulum termofilik maka perlu dipelajari proses start-up dengan inokulum mesofilik. Dalam penelitian ini dilakukan eksperimen menggunakan inokulum mesofilik waste activated sludge (WAS) dan substrat garbage slurry (GS) dengan tujuan berfokus pada strategi start up dan aklimatisasi mikroorganisme termofilik dari sumber mesofilik dengan menaikkan suhu secara langsung ke 55 oC dan menaikkan OLR secara bertahap. Strategi start up tersebut kemudian diaplikasikan pada eksperimen berikutnya yang bertujuan untuk mengevaluasi dan membandingkan kinerja tiga konfigurasi reaktor (fixed bed dengan media imobilisasi unwoven textile serta CSTR dan AFBR dengan media zeolit) terhadap populasi mikroorganisme dan produksi biogas (ditinjau dari laju pembentukan metana dan yield metana). Eksperimen strategi start up dengan variasi konsentrasi COD substrat (30.000 dan 15.000 mg/L) terbagi menjadi periode starvasi, propagasi, dan steady state. Hasil eksperimen menunjukkan pada periode starvasi yang berlangsung selama 20 hari telah ditemukan dominasi Methanothermobacter dan Methanosarcina sebagai mikroorganisme metanogen termofilik. Hal ini menunjukkan strategi start up single step efektif menseleksi mikroorganisme metanogen termofilik. Pada periode propagasi, reaktor dengan konsentrasi COD substrat tinggi menunjukkan proses hidrolisis berjalan lebih cepat dan laju pembentukan metana lebih tinggi. Hasil analisis sequence 16S rRNA menunjukkan terdapat perbedaan korelasi pada genus bakteri dengan parameter proses (OLR, laju pembentukan metana, dan individual VFA), namun mikroorganisme yang dominan memiliki peran yang sama baik di dalam reaktor dengan konsentrasi substrat tinggi maupun rendah. Pada periode steady state, bakteri asidogenik masih mendominasi, namun tidak ditemukan adanya akumulasi VFA. Hasil komparasi dari ketiga reaktor (fixed bed, CSTR dengan zeolit/ CSTR-Z, dan AFBR) menunjukkan bahwa media unwoven textile mampu mempertahankan kedekatan (proximity) mikroorganisme yang bersintropi, sehingga yield metana yang dihasilkan menjadi lebih tinggi yaitu 0,66 mL-CH4/g-sCODremoved. Pada reaktor CSTR-Z, media zeolit alam (tanpa pretreatment) belum optimal digunakan sebagai media imobilisasi. Selain itu, dengan adanya pengaduk mekanik dapat mengganggu perlekatan mikroorganisme pada zeolit, sehingga yield metana yang dihasilkan lebih rendah dari reaktor fixed bed yaitu 0,35 mL-CH4/g-sCODremoved. Pada reaktor AFBR aliran resirkulasi cairan di dalam reaktor dan media yang terfluidisasi dapat membantu kontak substrat dengan mikroorganisme pada media menjadi lebih sering, sehingga yield metana yang dihasilkan lebih tinggi (0,42 mL-CH4/g-sCODremoved) dari reaktor CSTR-Z dengan media yang sama. Dari hasil analisis sequence 16S rRNA kedua jenis media (unwoven textile dan zeolit), media unwoven textile lebih disukai mikroorganisme metanogen. Namun jenis mikroorganisme dominan yang ditemukan dari kedua reaktor (fixed bed dan CSTR dengan zeolit) relatif sama yaitu Haloplasma, Defluviitoga, dan Coprothermobacter sebagai bakteri hidrolitik, Lactobacillus sebagai bakteri asidogenik, Acetobacterium sebagai bakteri asetogenik, dan Methanothermobacter sebagai mikroorganisme metanogenik. Dari hasil simulasi menggunakan model ADM1 dengan perangkat lunak AQUASIM menunjukkan ketiga konfigurasi reaktor yang berbeda lebih memengaruhi nilai konstanta km daripada Ks, yang menunjukkan pengaruh konfigurasi reaktor cenderung lebih besar dalam meningkatkan kecepatan pertumbuhan dan jumlah mikroorganisme.

This study aims to increase biogas production through reactor configuration and microbial immobilization media. The immobilization media were added to enhance the population of microorganisms involved in the methane production from thermophilic anaerobic digestion of vinasse. The research was conducted in four aspects, namely: 1) start-up experiment, 2) fixed bed reactor, 3) continuous stirred tank reactor with zeolite (CSTR-Z), and 4) anaerobic fluidized bed reactor (AFBR). The drawback of the thermophilic process is the limited source of thermophilic inoculum; therefore, the first step of the study was appropriately acclimatizing the mesophilic inoculum to develop sufficient thermophilic microbial population. This experiment used waste-activated sludge (WAS) as mesophilic inoculum and garbage slurry (GS) as substrate to focus on the start-up strategy and enrichment of thermophilic microorganisms from mesophilic sources by increasing the temperature directly to 55 oC and increasing the OLR gradually. The start-up strategy was then applied to the next experiment to compare the performance of three reactor configurations (fixed bed with unwoven textile, CSTR and AFBR with zeolite as immobilization media) on the microbial population and biogas production (in terms of methane production rate and methane yield). The start-up strategy experiment with substrate COD concentration variations (i.e., 30,000 and 15,000 mg/L) was categorized into starvation, propagation, and steady-state periods. The experimental results showed that during the starvation period, which occurred for 20 days, the dominance of Methanothermobacter and Methanosarcina as thermophilic methanogenic microorganisms was found. The microbial composition confirmed that a single-step start-up strategy effectively selects thermophilic methanogenic microorganisms. In the propagation period, the reactor with a higher COD concentration showed a faster hydrolysis process and a higher methane production rate. The results of the 16S rRNA sequence observed different correlations between the types of bacteria with process parameters (OLR, methane production rate, and individual VFA). Still, the dominant microorganisms had the same function in the reactor with high and low substrate concentrations. In the steady state period, acidogenic bacteria still dominate, but no VFA accumulation was found. The comparison results of the three reactors (fixed bed, CSTR with zeolite/CSTR-Z, and AFBR) found that the unwoven textile media could maintain the proximity of syntrophic microorganisms. Therefore, the methane yield was higher, i.e., 0.66 mL-CH4/g-sCODremoved. In the CSTR-Z reactor, natural zeolite media (without pretreatment) was not optimally used as an immobilization media. In addition, the presence of a mechanical stirrer can interfere with the attachment of microorganisms to the zeolite, causing the methane yield to be lower than the fixed bed reactor, i.e., 0.35 mL-CH4/g-sCODremoved. In the AFBR, liquid recirculation and zeolite fluidization improved the contact between the substrate and the microorganisms on the media. Therefore, the methane yield is higher (0.42 mL-CH4/g-sCODremoved) than in the CSTR-Z reactor with the same media. From the analysis of 16S rRNA sequences of both types of media (unwoven textile and zeolite), unwoven textile media was preferred by methanogenic microorganisms. However, the dominant types of microorganisms in the fixed bed and CSTR with zeolite were relatively the same, which were Haloplasma, Defluviitoga, Coprothermobacter (hydrolytic), Lactobacillus (acidogenic), Acetobacterium (acetogenic), and Methanothermobacter (methanogenic). The simulation results using the ADM1 model with AQUASIM software showed that the different reactor configurations affected the value of the km constants more prominently than Ks. The different km values indicated different microbial growth rates in the three reactor configurations studied in this research.
